| Objectives To observe the effect and mechanism of Shenshao oral liquid preconditioning on myocardial ischemia reperfusion injury in rats.Methods All SD rats were randomly divided into 5 groups : the sham operation group,the ischemia reperfusion group,low dose of shenshao oral liquid group, high dose of shenshao oral liquid group and atorvastatin group. Low dose of shenshao oral liquid group, high dose of shenshao oral liquid group and atorvastatin group were given Shenshao oral liquid 4ml、8ml,atorvastatin 5.8mg Ig 3 days before operation, the sham operation group and the ischemia reperfusion group were given physiological saline. Use the method of ligaturing the left anterior descending coronary artery on rats 30 min and perfusing 120 min to build myocardial ischemia-reperfusion injury model,except the sham operation group.After making the model,all the rats will be put to death,then to extract the material. The expression of MMP-2 and TIMP-2 in myocardium,the myocardial enzyme levels, the left ventricleand infarction area and HE staining in myocardium on rats in each group were observed.Results 1 Observe the expressions of MMP-2 and TIMP-2 by immunohistochemistry:MMP-2:There is a very small amount expression in the sham operation group(5.24±1.74);Comparing with it, the ischemia reperfusion group increases significantly(82.07±10.04)(P<0.05);Comparing with the ischemia reperfusion group, high dose of shenshao oral liquid group decreases clearly most(22.22±6.57), low dose of shenshao oral liquid group(55.66±15.90) and atorvastatin group(50.55±6.12)are the second.TIMP-2:There is a very small amount expression in the sham operation group(73.79±5.65); There is no difference comparing with the ischemia reperfusion group(P>0.05);The least amount is high dose of shenshao oral liquid group(239.53±44.95);Low dose of shenshao oral liquid group(166.24±47.01)and atorvastatin group(149.03±16.66)are the second.2 Detect myocardial enzymes:Creatine phosphokinase-Mb: There is a very small amount expression in the sham operation group(74.81±16.16U/L);The ischemia reperfusion group increases significantly(6042.22±930.79U/L)(P<0.05);Comparing with the ischemia reperfusion group low dose of shenshao oral liquid group(3082.65±532.49U/L)and atorvastatin group(3235.3±547.22U/L)decrease clearly,and there is no difference between the two.High dose of shenshao oral liquid group decreases clearlymost(1475.3±621.27U/L).Lactate dehydrogenase: There is a very small amount expression in the sham operation group(48.17±9.89U/L);Comparing with it the ischemia reperfusion group increases significantly(2769.5±595.71U/L);LDH of other groups decrease obviously,low dose of shenshao oral liquid group(1833.25±525.81U/L), atorvastatin group(1524.5±592.48U/L),high dose of shenshao oral liquid group(915.71±207.16U/L),there is statistical significance among groups(P<0.05).3 Evaluate myocardial infarction area by EB-TTC double staining:There is no infarction in the sham operation group,the most infarction area is in the ischemia reperfusion group(74.39±1.73),the least is in high dose of shenshao oral liquid group(63.33±1.52),others are low dose of shenshao oral liquid group(67.24±1.98)and atorvastatin group(66.53±1.24).4 HE staining:The morphological structure of myocardial cell in the sham operation group is normal; the ischemia reperfusion group is broken badly;other groups are ameliorative,and high dose of shenshao oral liquid group is the best among these three group.Conclusions 1 Shenshao oral liquid or atorvastatin preconditioning both can restrain abnormal expression gain of MMP-2,increase the level of TIMP-2.At the same time,the two medicine can reduce the myocardial enzymes, the myocardial infarction area,and the damage of myocardial cell.So the two both have a protective effect on myocardium.High dose of shenshao oral liquid group is best in the matter of curative effect. 2 The mechanism of protection on Shenshao oral liquid may be related with reducing expression of MMP-2 and increasing expression of TIMP-2 in the early process of myocardial ischemia reperfusion by restisting oxidative stress. |
