Efects Of Nuodikang Capsule On Learning,memory And Expression Of ACHE And CHAT In Hippocampus Of AD Rats

Objective To explore the effect of Nuodikang capsule on learning and memory and activities of AchE and ChAT in hippocampus tissue of Alzheimer's disease(AD) animal model.Methods 32 rats were randomly divided into four groups as following,8 rats in each group: Aβ1-40 micro-injection group(model group), Aβ1-40 micro-injectiond and Aricept Tablets given group(Aricept Tablets group),Aβ1-40 micro-injectiond and Nuodikang capsule given group(Nuodikang capsule group )and normal group. The amyloidβ1-40(Aβ) was injected into the bilateral hippocampus to form AD model in rats,Morris water maze testing system was adopted to observe the changes of rats'. The appearance of AD rats hippocampal neurons were observed by HE staining. The activities of AchE and ChAT were detected by immunohistochemisty. The changes of AchE and ChAT mRNA were detected by RT-PCR.Results The escape latency of the place navigation in Aricept Tablets group(62.54±11.27s),Nuodikang capsule group(66.57±14.50s) and normal group(49.99±8.93s) were significantly shorter than which in the model group(86.46±5.81s)(P<0.05).The average optical density of positive AchE cells significantly reduced in Aricept Tablets group(0.32±0.05),Nuodikang capsule group(0.40±0.09), normal group(0.27±0.05), compared to model group(0.46±0.07)(P<0.05). The average optical density of positive ChAT cells significantly increased in Aricept Tablets group(0.23±0.02) ,Nuodikang capsule group(0.24±0.05), normal group(0.30±0.03), compared to model group(0.19±0.06)(P<0.05). The level of AchE mRNA significantly reduced, in Aricept Tablets group(0.29±0.05),Nuodikang capsule group(0.58±0.09), normal group(0.14±0.07), compared with model group(0.81±0.11)(P<0.05). The level of ChAT mRNA significantly increased in Aricept Tablets group(0.24±0.06) , Nuodikang capsule group(0.27±0.07), normal group(0.46±0.14), compared with model group(0.12±0.07)(P<0.05)Conclusion Nuodikang capsule can improve the ability of learning and memory in AD rats and the mechanism maybe is related to ChAT,AchE.
