Purpose:Legislation and prescription based on Replenishing kidney- essence and removing Phlegm and blood (Bu yuan Cong nao decoction) in the treatment of Alzheimer's disease (AD) model rats were observed on memory ability and brain cholinergic system.Material and method:Select SD strain male rats 75, 18 - 22w, were randomly divided into normal control group, sham operation group, model group,traditional Chinese medicine group, Western group 5 group, by injection in the hippocampus region Amanita grass acid (IBO ) andβamyloid peptide 1-42 (Aβ1-42) causing the mixture AD model was established 2 weeks after recovery to complement the Bu yuan Cong nao decoction and medicine decoction suspension administered pira- cetam treatment, continuous 28 days. After the administration, through behaveio- ral (Morris water maze) test, biochemistry (choline acetyl transferase ChAT, acetyl cholinesterase activity of AchE) colorimetric assay and morphological changes and positive cells in hippocampus of Aβexpression observed, clear up Bu yuan Cong nao decoction AD model rats on behavior and brain cholinergic system in rats.Results:1. Behavior: Morris water maze results, the normal group and sham operation group little difference; model group, the average incubation period than the normal group, sham operation group was significantly longer (P<0.01); Western medicine group, traditional Chinese medicine group, the average incubation period was significantly reduced compared with the model , significantly different (P<0.01); Chinese medicine and western medicine group was no significant difference (P> 0.05).2. Morphological changes: The model shows that a large number of rat hippocampus neurons necrosis, cell shrinkage, condensation nuclei, chromatin deeper,irregu- lar in shape and strong expression of Aβ. Treatment group were improved. 3. Biochemical indicators: for ChAT and AchE activity of the normal group and sham operation group little difference; with the normal group, model group was significantly lower (P <0.05); Compared with model group, the two treatment groups increased to varying degrees (P <0.05); for ChAT activity between the two treatment groups was compared, the difference is small (P> 0.05); for AchE activity traditional Chinese medicine group was significantly better than the western medicine group (P <0.05).Conclusion:1. Aβ1 -42 + IBO rats with damaged hippocampus of rat models to establish AD, AD is more close to the simulation of the clinical manifestations and basic pathology, is a reliable animal model.2. Bu yuan Cong nao decoction can improve the AD model rat brain ChAT and AChE activity levels, repair, cholinergic neurons, with the improvement of impaired cholinergic system function.3. Bu yuan Cong nao decoction can improve the pathological changes in hippocampus and reduce Aβexpression, reducing neurotoxic injury.4. Bu yuan Cong nao decoction up on the AD model rats significantly improved learning and memory function.
