Objective:Detecting the expression of Gli1 and VEGF in 78 cases of colorectal cancer and 30 cases of corresponding normal colorectal tissue .To explore the relationship between the expressions of Gli1 and vascular endothelial growth factor(VEGF)in colorectal cancer and their clinicopathologic features,and their role in the development and progression of colorectal cancer.Methods: Specimens of 78 patients with colorectal cancer and 30 cases of corresponding normal tissue were choosed from the department of gastrointestinal surgery in the first affiliated hospital,chongqing medical university .The expression of Gli1 and VEGF was assessed by immunohistochemistry and Western blot, and its correlation with clinicopathological factors was statistically analyzed to investigate the effect of Gli1 and VEGF proteins on colorectal cancer.Results : Gli1 and VEGF have a higher expression rate in colorectal cancer, they expressed in nucleus and cytoplasm respectively. The expression of Gli1 and VEGF in colorectal cancer had no correlation to patients,age, tumor size, pathologic type(P>0.05), but the expression of VEGF and Gli1 were correlated to lymph node metastasis and Duke stage (P<0.05).The expression activity of Gli1 and VEGF in Dukes C+D was significantly higher than that of Dukes A+B(P<0.05).they express highly in the group of colorectal cancer with lymphatic and distant metastasis than the group without metastasis. Gli1 expression was positively correlated to VEGF expression (r=0.865,P<0.05).Conclusion: Gli1 and VEGF have collaborative and mutual regulation in the development of colorectal cancer, the Gli1 and VEGF expression participate in the invasion and metastasis process of colorectal cancer. Their joint detection can be looked upon as an important symbol for judging the prognosis of colorectal cancer , screening the high-risk metastasis patients and guiding the therapy of colorectal carcinoma.
