ObjectiveTo detect the expressions of Survivin,fragile histidine triad(FHIT) and vascular endothelial growth factor C(VEGF-C) in gastric adenocarcinoma, investigate the correlation among the three factors and their important roles and possible mechanism in the progress of originating and developing of human gastric adenocarcinoma.MethodsApplying immunohistochemical method to detect the protein expression of Survivin,FHIT,VEGF-C in 43 cases of gastric adenocarcinoma, the corresponding tumor-adjacent tissues ,distal normal tissues, and compare the differences of their expressions in three groups and analyzing their expressions in gastric adenocarcinoma and the relationships with clinical pathology;Survivin,FHIT,VEGF-C mRNA expressions were detected by Semi-quantitative RT - PCR, and the differences of their expressions in three groups were compared , and their relationships with clinical pathology were analyzed.ResultsIn gastric adenocarcinoma organization ,the protein and mRNA expressions of Survivin, VEGF-C were significantly higher than the corresponding tumor-adjacent tissues ,distal normal tissues, FHIT protein and mRNA expressions were significantly lower than tumor-adjacent tissues ,distal normal tissues (P<0.05). the expressions of Survivin, VEGF-C protein and mRNA in gastric adenocarcinoma were connected with tumor differentiation, stages, infiltration, lymph node metastasis (P<0.05). the expressions of FHIT mRNA and protein and in gastric adenocarcinoma were connected with tumor diameter, stages, infiltration, lymph node metastasis (P<0.05). In gastric adenocarcinoma the protein expressions of survivin and VEGF-C were positively correlated (r=0.656, P<0.01), and the protein expressions of Survivin and FHIT were negatively correlated (r=-0.621, P<0.01). In gastric adenocarcinoma the mRNA expressions of survivin and VEGF-C were positively correlated (r=0.675,P<0.01), and the mRNA expressions of survivin and FHIT were negatively correlated (r=-0.638,P<0.01)ConclusionIn the protein level and mRNA level, Higher expressions of Survivin and VEGF-C and lower expression of FHIT collaboratively promote occurrence and development of gastric adenocarcinoma.
