Objective:To observe and compare the therapeutic efficacy and the transdermal performance of old and new formulations Xiaoyusan in external use of treating New Zealand White rabbit models with acute injury of soft tissue, namely acute blood stasis, and to select the advantage formulation.Methods:60 New Zealand White rabbits were attacked to obtain animal models of blood stasis, and randomly divided into 4 groups:A.model group, treated with empty one; B.ointment group, treated with the old formulation; C.cataplasm group, treated with the new formulation; D. wet compress patch group, treated the same as C. Each group was treated respectively with different formulations of Xiaoyusan for 2,4 and 6 days, general observation were taken and scored; HE were stained histological observation under light microscope; plasma concerntration (emodin) were determined by the high performance liquid chromatographic (HPLC); guoup B and group C in vitro penetration test with the in vitro rabbit skin and the Franz the diffusion cell, then analyzed and compared. Results:There was difference in general observation comparing between group D and group B in day 4, group C and group B in day 6 (P<0.05, P<0.01); HE staining show that group D excelled group B in day 4 in grades of pathological tissue section (P<0.01); blood concentration showed that the differernce of the new formulation groups (group C and group D) and the old formulation group (group B) was statistically significant in 2,4,6 days (P<0.01); in citro penetration test illustrates, group C emodin in vitro transmittance and total volume is higher than the group B.Conclusion:Xiaoyusan new formulation in the external use of treating the New Zealand White rabbit models with acute blood stasis was better than old formulation in absorbability and curative effect. Comparison between two new foumulations is no statistically diference (P>0.05), but wet compress patch group is better than cataplasm group in specific points.
