Objective:Observation and comparison of old and new dosage forms of Xiaoyusan (plastic, In-situ gel gel and powder) local external application on acute soft tissue injury of New Zealand White rabbit model of treatment effects and percutaneous absorption, selective advantage of dosage forms, and to investigate its promotion of acute soft tissue contusion repair mechanism.Methods:60New Zealand White rabbits caused by acute soft tissue contusion model, were randomly divided into powder group (A), plastic group (B), In-situ gel gel group (C), model group (D), A、 B group and C group respectively Xiaoyusan coating agent, namely type gel and powder in treatment of, group D with blank patch application. Each group after administration of2D,4D,6D for gross observation scale; each group with5rats of experimental rabbits, in accordance with the requirements of sample processing and HE staining under the light microscope to observe the histological changes; A, B group and C group with the modified Franz diffusion cell and isolated rabbit in vitro transdermal experiment device through the statistical analysis, comparison of various forms of penetration rate, cumulative transdermal percentage with various skin reserves difference.Results:Gross observation score, C group and A group4D,6D, B group and A group6D, local swelling, bruising and other symptoms relatively before have improved significantly (P<0.01); optical microscope (10×10) was observed in HE staining, C group6D histology score was better than A group (P<0.05); in vitro transdermal characteristics show transdermal speed from fast to slow, skin reserves from high to low, transdermal aggregates from at least as followed:the alcohol extract>> powder aqueous extract, alcohol extract of transdermal speed is the quickest, most skin reserves, transdermal most gross; various forms. The in vitro transdermal experiment results show:the gel transdermal speed is the quickest, most skin reserves, plastic transdermal most gross.Conclusion:Xiaoyusan external use in treating acute soft tissue contusion in New Zealand White rabbits model, new formulations of the absorption and efficacy is superior to older forms of Xiaoyusan, and coating agent and gel two new forms of no significant difference (P>0.05), whereby the gel in the treatment of acute soft tissue contusion effects in terms of overall slightly better than the film agent.
