Objective:we establish the model of devilery trauma to simulate the human pregnancy, delivery and devilery trauma, use the low-decibel infrasound as the treatment factor, and then observe the effect of low- decibel infrasound on CollagenⅠand CollagenⅢin vaginalwall and the levels of estrogen and progesterone of rats.Materials and methods:1. Animal grouping:Assign 8 rats from the 30 female rats randomly as blank control group (A group) for routine feeding and the remaining female rats were caged together with male rats in the ratio of 2:1. One month later,16 female rats were pregnant. After establishing the model of delivery trauma, they were randomly divided into two groups, the negative control group (B group) with no treatment and the experimental group (C group) with infrasound treatment.2. Establishing delivery trauma models:After their delivery, female rats were immediately injected intraperitoneally with 10% chloral hydrate (3mL/kg) for anesthesia and fixed in a supine position; their vulva were disinfected, the epidural catheters were inserted into the urinary tract to empty the bladder,14F catheters were inserted about 3cm into the vagina,5mL saline was injected into the foley for vagina distraction; 1# wire was fixed to the vaginal orifice; the rats were unfixed to remain a natural extension for 4h; after the catheters were removed, the rats were fed in the cages.3. Infrasound parameters and treatment:Infrasound therapeutic apparatus infratonic 8000 is mainly composed of by the host machine and the sound head in a frequency spectrum of 2~25Hz. The main frequency spectrum of infrasound changes randomly and the sound pressure level is 70dB. Fixed the rats by the rat holder and held the sound head vertically to the horizontal position of the vagina of rats. The shooting head and the rat vagina were in a distance of about 2~3cm. After establishing the model, the experimental group were given infrasound treatment 30 mins each time, twice a day, last for 20 days. 4. Detection:20 days after, anesthetized rats and obtained the materials from the vaginalwall. Collagen I and Collagen III were detected by immunohistochemistry and used Leica Qwin Plus image analysis system to calculate the area percentage of collagen I and Collagen III in order to obtained their relative content, the change in morphology of vaginalwall in each group was observed with HE staining and Masson staining. Used ELISA (enzyme linked immunosorbent assay) and RIA (radioimmunoassay) to test the levels of E2, P, FSH, LH, PRL in blood serum of rats.5. Statistics:All data were shown in x±s and treated with SPSS13.0 software in statistics.Independent-samples t-test was used in comparison between groups for analysis. P<0.05 showed significant differences.Result:1. The effect of low-decibel infrasound on Collagen I and Collagen III in vaginalwall of rats: The mean value (%) of relative area of collagen I and collagen III in vaginalwall of the blank control group respectively was 11.43±2.67,1.85±0.44;The mean value (%) of relative area of collagenⅠand collagenⅢin vaginalwall of the negative control group respectively was 3.68±1.04,0.60±0.15; The mean value (%) of relative area of collagen I and collagenⅢin vaginalwall of the experimental group respectively is 8.17±1.06,1.32±0.23; The content of collagenⅠand collagenⅢin the vaginalwall of the blank control group was significantly higher than that in the negative control group (P<0.05); The content of collagenⅠand collagenⅢin the vaginalwall of the experimental group was significantly higher than that in the negative control group (P<0.05); The content of collagenⅠand collagenⅢin the vaginalwall of the blank control group was significantly higher than that in the experimental group (P<0.05).2. The effect of low-decibel infrasound on the ovarian function of rats:The levels of E2, P, FSH, LH, PRL in blood serum of the blank control group respectively were 12.57±2.57(ng/mL),14.27±3.57(ng/mL),15.72±2.04(mIU/mL),9.97±1.40(mIU/mL), 11.72±4.84(ng/mL); The level of E2, P, FSH, LH, PRL in blood serum of the negative control group respectively was 7.75±3.33(ng/mL),10.07±0.95(ng/mL),14.06±1.51 (mIU/mL),10.89±3.46(mIU/mL),11.70±4.22(ng/mL);The level of E2, P, FSH, LH, PRL in blood serum of the experimental group respectively was 8.37±2.83(ng/mL),10.20±1.93 (ng/mL),13.79±1.60(mIU/mL),8.93±1.60(mIU/mL),9.24±2.04(ng/mL). The level of E2 and P of the blank control group was significantly higher than the negative control group and the experimental group (P<0.05); The level of E2 and P had no significantly difference between the negative control group and the experimental group (P>0.05); The level of FSH, LH, PRL had no significantly difference among these three groups (P>0.05).Conclusion:1. Pregnancy, delivery and delivery trauma reduce the content of collagenⅠand collagenⅢin vaginalwall of rats2. Low-decibel infrasound can increase the content of collagenⅠand collagenⅢin vaginalwall of rats after delivery trauma3. Low-decibel infrasound can increase the content of collagenⅠand collagenⅢin vaginalwall of rats after delivery trauma, but the level of them is still lower than that in unpregant rats.4. Pregnancy, delivery and delivery trauma reduce the levels of E2 and P in the rats'serum, but it does not change the levels of FSH and LH.So postpartum rats' E2 and P may be in the bottom of normal range referral.5. The low-decibel infrasound does not improve the reduction of E2 and P which was caused by pregnancy, delivery and delivery trauma of rats.
