Objective:To observe the inhibition of Compound Guizhu Capsule (CGZC) and synergy combined with chemotherapy drugs on the growth of mice Lewis lung cancer tissue and the effect on the expression of PTTG gene and PCNA of tumor tissue, and to study the mechanism of which to provide the theory basis for clinical application of CGZC.Methods:Modeled the mice Lewis lung carcinoma through inoculating Lewis lung carcinoma cell lines into mice. Weighed them after 24 hours, and randomly divided them into 8 groups of Tumor-burdened model control group, DDP group, the group of high dose of CGZC, the group of middle dose of CGZC, the group of low dose of CGZC, the group of high dose of CGZC with DDP, the group of middle dose of CGZC with DDP, the group of low dose of CGZC with DDP. There were 10 rats in each group, male and female in equal. At the 24th hours after inoculating, medicine were given,0.4 ml distilled water was perfused into stomachs of Tumor-burdened model control group,1 mg DDP for each kilogram mouse was injected into abdominal cavity once a day for 3 days. CGZC were diluted with distilled Water and perfused into stomachs based on mice's weight once every day for 10 days,20g diluents for each kilogram mouse (10 times more dosage than adults')for the group of high dose of CGZC,10g(5 times more dosage than adults')for the group of high dose of CGZC and 5g(2.5 times more dosage than adults') for the group of high dose of CGZC. At the 24th hours after the last perfusing, weighed the mice, and then killed them and weigh the mice's tumor, lung, spleen, thymus, adrenal gland. Calculated weight, the indexes of spleen, adrenal gland and thymus and inhibition rates for tumor's growth and metastasis, and tested expression of the gene of PTTG and PCNA by immunohistochemical method.Results:CGZC can remarkablely inhibit the growth of Lewis lung tumor (P<0.01),inhibition of high dose group was 39.83%,37.57% in the dose group and low dose group 31.85%; and the inhibition rates with CGZC and DDP is higher than with DDP only (P<0.01), high dose group was 67.28% inhibition,65.23% in the dose group and low dose group 63.10%. CGZC could significantly improve the immune organs index, and the CGZC high dose group's effect is the most significant. (Compared with the control group, P<0.05) CGZC could reduce expression of PCNA and PTTG protein of the Lewis lung tumor tissue. (Compared with the control group, P <0.05). And CGZC inhibited the number of lung metastases of Lewis lung carcinoma in mice, which was statistically significant (Compared with the control group, P<0.05).Conclusion:CGZC could remarkablely inhibit the growth of mouse Lewis lung cancer tissue, and have synergistic inhibitory effect on mice Lewis lung cancer, combined with chemotherapy drugs, the mechanism of which may be related to reducing the expression activity of PTTG gene and PCNA. CGZC can inhibit spontaneous lung metastasis of Lewis lung cancer in mice; CGZC have significant enhancement and protection effect on the immune function of Lewis lung cancer mice, and can inhibit the spontaneous metastasis of mouse Lewis lung cancer.
