Objective: To study the expression of DDR1 and bFGF at different times of rat kidney with tubulointersititial fibrosis, as well as the effect of genistein on renal tubular injury,interstitialfibrosis and expression of DDR1 and bFGF.Methods:Forty-five healthy male Wistar rats weighing 180-220g were divided into 3 groups:group 1,sham-operated group(sham group,n=15);group 2, unilateral ureteral obstruct group(UUO group,n=15);group 3,genistein group(Gen group,n=15).Before 1 day of operation,Sham group and UUO group were poured with 1ml NS every day,Gen group were poured with genistein 30mg-kg-1·d-1.UUO group and Gen group were established renal tubulointersititial fibrosis model by ligation the unilateral ureteral of rats.Sham group were only exposed ureter,but not ligation the ureteral of rats.The kidney tissues were collected after 7,14,21 days treatment.The level of tubulointerstitial fibrosis was observed by HE and Masson staining.The protein expression of DDR1 and bFGF in kidney tissue were measured by immunohistochemistry.Result:Compared with Sham group, the expression of DDR1,bFGF increased obviously in UUO group (P<0.05).Compared with UUO group, the renal tubule damage and tubulointerstitial fibrosis had been reduced, also the expression of DDR1,bFGF decreased significantly in genistein group (P<0.05).Conclusion:The expression of DDR1 and bFGF accord with renal tubule damage and tubulointerstitial fibrosis and fibrosis.Genistein could alleviate renal tubule damage and tubulointerstitial fibrosis through downregulation DDR1,bFGF expression possibly.
