Objective: By detecting the expression of Id-1, Id-3, MMP-9 in brain metastatic tumors, and to explore their role in the process of formation and the invasion of metastases.Materials and methods:1 To collect the samples of brain metastatic tumors by surgical resection from 44 cases and samples of normal brain tissue 10 cases(normal brain tissues by surgical resection from interior decompression in patients with cerebral hern- ia). All of the tissues were fixed in 10% neutral formalin, embedded in paraff- in generally and cut into slices serially. To retrieve corresponding patients with primary tumor paraffin blocks of tumor specimens archived in pathology, cut into slices serially.2 Set of brain metastatic tumor and primary tumor for the study group and normal brain tissue as the control group. And according to age, gender or primary tumor site set the control group respectively. The detection of MMP-9, Id-1, Id-3 was made of using immunohistochemical Streptavidin-Persidase (SP) technique in brain metastatic tumor, primary tumor and normal brain tissue. Each section at high magnification (×400) count 10 high power field, according to the percentage of positive cells assessed positive rating, to calculated the percentage of positive cells.3 Collect clinical pathology material of all the samples. Processing the acquired data and analyzing the correlation between MMP-9, Id-1, Id-3 in brain metastatic tumor with SPSS13.0 statistic software.Results:1 The positive rates of Id-1, Id-3, MMP-9 expression in 44 cases of brain metastatic tumor were 93.2%(41/44),95.5%(42/44),81.8%(36/44) respectively. The positive rates of Id-1, Id-3, MMP-9 expression were 71.4% (5/7),71.4%(5/7),57.1%(4/7) in 7 cases of primary tumor archived in pathology. There was no expression of these three proteins in normal brain tissue.2 The expression of Id-1, Id-3, MMP-9 in brain metastatic tumor and primary tumor were all significantly higher than normal brain tissue(P<0.05).The difference between brain metastatic tumor and primary tumor was no statistical significance in expression of Id-1, Id-3, MMP-9(P>0.05). Based on age, gender and primary tumor site were different groups, Id-1, Id-3, MMP-9 in the different groups in the expression of brain metastatic tumor was no statistical significance(P>0.05).3 The correlation analysis of Id-1, Id-3, MMP-9 expression for all the brain metastatic tumor samples showed that there was a significant positive correlat ion among Id-1, Id-3 and MMP-9.Id-1 and MMP-9 expression is a positive correlation(γ=0.386,P=0.010), Id-3 and MMP-9 expression is a positive cor- relation(γ=0.306,P=0.043),Id-1 and Id-3 expression is a positive correlation(γ=0.319,P=0.035).Conclusion: Id-1, Id-3 and MMP-9 were over expressed in brain metast- atic tumor and primary tumor, and they were not expressed in normal brain tissue. The over expression of Id-1, Id-3 and MMP-9 in brain metastatic tumor was not associated with primary tumor site, age and gender. There was a signi- ficant positive correlation among Id-1, Id-3 and MMP-9, and the correlation between Id-1 and MMP-9 was most significant. It was suggest that Id-1 and Id-3 involved in the formation of brain metastatic tumor. And they adjust the over expression of MMP-9, to provide condition for the formation of metas- tatic tumor. The over expression of MMP-9, Id-1, Id-3 played an important biological function for the formation of brain metastatic tumor, and the three proteins had a role of synergies regulatory. This finding might put forward the mechanism of brain metastases.
