Objective: To detect the expression of OPN, CD44v6in brain metastatictumors, and to explore their role in the process of formation and the invasionof metastases.Materials and methods:1To collect the samples of brain metastatic tumors by surgical resectionfrom50cases and samples of normal brain tissue10cases (by surgicalresection from interior decompression in patients with cerebral hernia ofcraniocerebral injury). All of the tissues were fixed in10%neutral formalin,embedded in paraffin generally and cut into slices serially. Retrievecorresponding primary tumor paraffin blocks from pathology department, andcut into slices serially.2Set brain metastatic tumor and primary tumor as the study group andnormal brain tissue as the control group. And according to age, gender andprimary tumor site, the cases were divided into different groups respectively.The detection of OPN and CD44v6was made by immunohistochemicalStreptavidin-Persidase (SP) technique in brain metastatic tumor, primary tumorand normal brain tissue. Ten high power fields were chosen at highmagnification (×400) from every section, to calculate the positive rate basedon the percentage of positive cells.3Collecting clinical pathology date of all the samples. Processing theacquired data and analyzing the correlation between OPN and CD44v6inbrain metastatic tumor with SPSS13.0statistic software.Results:1The positive rates of OPN and CD44v6expression in50cases of brainmetastatic tumor were92%(46/50),88%(44/50)respectively. The positiverates of OPN and CD44v6expression were70%(7/10)and60%(6/10)in10 cases of primary tumor archived in pathology. There was no expression ofthese proteins in normal brain tissue.2The expression of OPN and CD44v6in brain metastatic tumor andprimary tumor were both significantly higher than normal brain tissue(P<0.05). Both of OPN and CD44v6express in brain metastatic tumor werehigher than primary tumor, but there were no statistical significance. Thedifference between OPN and CD44v6in brain metastatic tumor and primarytumor was no statistical significance(P>0.05). Based on age, gender andprimary tumor site were different groups, OPN and CD44v6in the differentgroups were no statistical significance(P>0.05).3The correlation analysis of OPN and CD44v6expression in brainmetastatic tumor samples showed that there was a significant positive correla-tion between OPN and CD44v6.Conclusion: The expression of OPN and CD44v6in brain metastatictumor and primary tumor were all significantly higher than normal brain tissue(P<0.05),both of OPN and CD44v6express in brain metastatic tumor werehigher than primary tumor,and they were not expressed in normal braintissue.The over expression of OPN and CD44v6in brain metastatic tumor wasnot associated with primary tumor site, age and gender. There was a signi-ficant positive correlation between OPN and CD44v6, It was suggested thatOPN and CD44v6involved in the formation of brain metastatic tumor. Theover expression of OPN and CD44v6played an important biological functionfor the formation of brain metastatic tumor, and they maybe have a role ofsynergies regulatory.
