Objective: To research the relations between hepatocytes apoptosis and expressions of Bax/Bcl-2 in rats with obstructive jaundice, and explore the effects of Prostaglandin E1 on those.Method: A total of 72 adult SD rats were randomly allocated to three groups: the sham operation group (CG group), the OJ group (OJ group) and the group of OJ rats administered Prostaglandin E1 (PE group). The OJ and PE group were established by double-ligating bile duct (BDL). Tail vein injection of Prostaglandin E1 was only administered daily in PE group, postoperatvely. Some rats in three groups were sacrificed on the 3th, 7th and 10th day of postoperation randomly (n=8). The levels of serum total bilirubin (TBil), direct bilirubin (DBil), alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were tested and the tissue's histopathology changes were observed;Apoptosis of hepatocytes was tested by TUNEL and accounted apoptosis index (AI), the immunehistochemical technique was used for detecting expressions of the Bax and Bcl-2 protein.Resultâ’ˆWith the time of BDL extending, values of serum ALT, AST, TBil and DBil were increased and values of OJ group and PE group were higher than those of CG group.â’‰With the time extending, the Bax and Bcl-2 protein of both OJ and PE group were increased following that cell apoptosis increased apparently in hepatic tissue. Compared with OJ group, the injury degree of hepatic function and histopathology of changes in PE group were decreased, The Bcl-2 protein of PE group was increased more evidently, just like Bax in OJ group.Conclusionâ’ˆObstructive jaundice in rats may lead to elevated levels of serum transaminase and bilirubin, hepatic lobule structural damage, increase of liver cell apoptosis and levels of Bax/Bcl-2 increased.â’‰PGE1 can ease the degree of hepatic functional injury in rats with obstructive jaundice, for it may decrease cell apoptosis by up-regulating the expression of Bcl-2 and down-regulating the expression of Bax in hepatic tissue.
