Purpose:First to detect the single nucleotide polymorphisms (SNPs) of the human T-cell immunoglobulin and mucin domain-3 (Tim-3) gene in the Uigurs of Xinjiang Uygur Autonomous Region and to search for the genetic marks of the Tim-3 gene; Then to find the the relationship between the polymorphism of the TIM-3 gene mononucleotide site rs10053538, rs13170556, rs4704853, rs6555849, rs10515746 together with NEW-SNP and the allergic rhinitis. Method:First employing the method of direct sequencing PCR amplified to detect the length of 5'promoter region 3kb and 3'ext 1kb of the Tim-3 gene to sequence the SNPs of 32 common Uigurs of Xinjiang, in order to institute the position, type and frequency of Tim-3 gene of Uigurs of Xinjiang.Then analyzed the six mononucleotide polymorphism site of 150 Uygur and Han AR patients and normal people respectively in Xinjiang in terms of SNaPshot SNP typing technique, after this calculate their gene type and alleomorph frequency. Then according to the TIM-3 gene layout, the gene type and the allelomorph frequency had been calculated. The Hardy-Weinberg balance law had been used for exam.and the LD analyses and haplotype frequencies had been calculated. Result:12 SNPs were detected in the Tim-3 gene, including 8 existing high-frequency SNPs and 4 newly-discovered low-frequency SNPs. The genotype and alleles of each frequency of SNPs were proved to attain heredity balance through the Hardy-Weinberg equilibrium law.Then polymorphism of the Tim-3 gene mononucleotide in the Uygur and Han Allergic rhinitis patients and normal people Xinjiang, in the Uygur, Compared with healthy group, the difference of these gene types layout and the allelomorph frequency has no statistic meaning (P>0.05), in the Han, Compared with healthy group, the difference of these gene types layout and the allelomorph frequency has no statistic meaning (P>0.05). There is no correlation between AR susceptibility and Tim-3 gene mononucleotide site rs10053538 rs13170556, rs4704853, rs6555849, rs10515746 and NEW-SNP of Uygur people and Han people in Xinjiang. The alleles of both healthy controls and AR cases have achieved genetic equilibrium in the four SNP sits. Conclusion:The SNP distribution of Tim-3 gene of Uigurs of Xinjiang reflects racial and municipal difference which can lay the foundation of study of the relativity between Tim-3 gene and disease on Uigurs and the Hans. And there is no correlation between AR susceptibility and Tim-3 gene mononucleotide site rs10053538 rs13170556, rs4704853, rs6555849, rs10515746 and NEW-SNP of Uygur people and Han people in Xinjiang.
