Objective To investigate four single nucleotide polymorphism sites of T cellimmunoglobulin domain and mucin domain protein-3(Tim-3) gene and susceptibilityof rheumatoid arthritis (RA) in QingHai Han population and Tibetan and providetheory basis for taking precautions.Methods Polymerase chain reaction and restriction fragment length polymorphism(PCR-RFLP) was carried out to examine the SNPs at-574,-882,-1541and4259sitesof Tim-3gene, including111healthy individuals and101RA patients in QingHaiHan population,102healthy individuals and60RA patients in QingHai Tibetan. Thegenotype and alleles frequencies were calculated and analyzed.Results①Both genotypic frequency and allelic frequency at-882and-574sites ofTim-3gene had no significant difference (P>0.01), while both genotypic frequencyand allelic frequency at-1541and4259sites had significant difference betweenpatient group and normal group (P<0.05) in QingHai Han population.②Bothgenotypic frequency and allelic frequency at-882、-574、-1541and4259sites ofTim-3gene had no significant difference (P>0.05) between patient group and normalgroup (P>0.05) in QingHai Tibetan.③Both genotypic frequency and allelicfrequency at-882and-574sites of Tim-3gene had no significant difference (P>0.05),while both genotypic frequency and allelic frequency at-1541and4259sites hadsignificant difference between Tibetan and Han population (P<0.01) in QingHaiHealthy population.④Both genotypic frequency and allelic frequency at-882、-574、-1541and4259sites of Tim-3gene had no significant difference between Tibetan andHan population (P>0.05) in QingHai patients with RA.Conclusion-1541and4259sites of Tim-3gene probably may have SNP variations in QingHai Han population and Tibetan. And-1541,4259mutation gene maybeassociate with RA in QingHai Han population and Tibetan.
