Study On The Anti-Microbial Activity Of Bamboo Vinegar

The bamboo vinegar produced in Suichang was tested for anti-microbial activities against six species of microbe, which included Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Saccharomyces cervisiae, Canidia albicans and Aspergillus niger. Bamboo vinegar was also tested for its stability to acid, alkali, temperature, ultraviolet and metallic ion. Morever, toxicological safety of bamboo vinegar was evaluated. The main components of the bamboo vinegar were also tested for anti-microbial activities against six species of microbe in order to determine the main active ingredients of bamboo vinegar.The simple refinement and the distill refinement of bamboo vinegar had been optimized and compared. Meanwhile, the surface bacteriostatic agent made of bamboo vinegar was prepared preliminary. The main results were summarized as follows:1.The bamboo vinegar has broad antibacterial spectrum,it was showed some inhibitory effect when the bamboo vinegar was tested on the six species of microbe. The minimal inhibitory concentration (MIC) of bamboo vinegar to Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa was 7.813μg/mL,to Saccharomyces cervisiae, Canidia albicans and Aspergillus niger was 62.5μg/mL. The minimal bactericidal concentration (MBC) of bamboo vinegar to Escherichia coli and Staphylococcus aureus was 31.25μg /mL,to Pseudomonas aeruginosa was 15.63μg/mL,The minimal bactericidal concentration (MBC) of bamboo vinegar to Saccharomyces cervisiae and Canidia albicans was 125μg/mL, to Aspergillus niger was 250μg/mL.The antimicrobial properties of bamboo vinegar was stable to temperature and metallic ion. After treated with alkaline, the bamboo vinegar showed loss of antibacterial activity. The UV radiation showed little effect on bamboo vinegar antimicrobial activity. In toxicological safety evaluationon on bamboo vinegar, the results showed that the bamboo vinegar was nonpoisonous in reality with oral LD50 more than 20mL·kg-1BW, while with intraperitoneal injection LD50 less than 10mL·kg-1BW. The bamboo vinegar had no skin irritation.3.Acetic acid and phenol in bamboo vinegar play an important role in anti-micrbial activity. Acetic acid was one of the main components of bamboo vinegar.The highest concentration of bamboo vinegar was acetic acid, the acetic acid in bamboo vinegar showed that it had certain anti-micrbial activity,its inhibition on bacteria was better than fungi.The concentration of phenol in bamboo vinegar was next to acetic acid,while its inhibition on fungi was better than bacteria. 2-Methoxyphenol and 2,6-Dimethoxyphenol also showed some anti-micrbial activity in bamboo vinegar. The concentration of 2-furaldehyde in bamboo vinegar almost had no anti-micrbial activity.4.Active carbon powder was chosen as a medium of the simple bamboo vinegar refinement.The best technological process of simple refinement was as follows: added 50g/L active carbon powder into bamboo vinegar,at room temperature and stirring the solution for one hour, then, filtered to obatained the refined bamboo vinegar. The best technological process of distill refinement was as follows: distilled the bamboo vinegar under vacuum 0.09Mpa and at 60℃~70℃,distillation volume was 90% of original bamboo vinegar.5.The bamboo vinegar surface bacteriostatic agent prepared as follows: distilled the bamboo vinegar under vacuum 0.09Mpa and at 60℃~70℃,distillation volume was 90% of original bamboo vinegar,then to 500:1 (volume ratio) added the citrus essential oil into refined bamboo vinegar. The bamboo vinegar surface bacteriostatic agent had no skin irritation and with intraperitoneal injection LD50 about 10mL·kg-1BW.
