Objective:To investigate the role of adiponectin and nuclear factor kappa B p65 (NF-κBp65)on metastasis and invasion in colorectal carcinoma by observing the expression of adiponectin and NF-κBp65 in colorectal carcinoma and their relationship with clinical pathologic features.Methods:Immunohistochemical staining was performed to detect the expression of adiponectin and NF-κBp65 in colorectal carcinoma (n = 42), colorectal adenoma (n = 20) and corresponding normal controls (n = 42). Image analysis software was used for semi-quantitative assessment and analysis their relevance.Results:In colorectal carcinoma and colorectal adenoma group, adiponectin expressed small in cell envelope and cytolymph of tumor cell. And in normal colon group, adiponectin expressed in envelope and cytolymph of glandular cell of colon tissue equably. The integrated optical densities and the positive expression rate of adiponectin was increased progressively from cancer tissue,colorectal adenoma and normal controls [40.5%,75%,85.7%;(15.35±3.81),( 38.71±6.80),( 43.03±4.89)]. The difference between normal controls and adenoma to cancer tissue was significant ( P < 0.05). The integrated optical densities of NF-κBp65 was decreased progressively from normal controls, colorectal adenoma and cancer tissue[78.6%,50%,16.7%; (34.43±2.36),( 27.15±6.11),(15.70±2.36), P <0.05].The expression of adiponectin and NF-κBp65 in colorectal carcinoma was associated with the loss of tumor differentiation, metastasis and TNM stage ( P < 0.05). There was a significance negative correlation between adiponectin and NF-κBp65.(r=-0.881,P<0.01). Conclusions:Adiponectin is lowly expressed in colorectal carcinoma,but NF-κBp65 is highly expressed..Adiponectin may play an essential role in colorectal carcinoma by the NF-κBp65 pathways.
