Background:With neoadjuvant chemotherapy in clinical treatment of patients with cervical cancer,the overall survival have elevated,but traditional cases have series of toxicity, seeking new safe and effective drugs has become a research hotspot at home and abroad in recent years. Traditional Chinese drugs have the characteristics of little side effects. Recent researches show that allicin can enhance the effective antitumor function,and can reduce the side effects,but its mechanism is not clear.Objective:To discusses if allicin can enhance the drug efficacy through studying the change of the proliferation and apoptosis of human cervical cancer cells Hela after the treatment of allicin combinated with oxaliplatin,and to further explore the possible mechanism and provides theoretical basis for clinical application.Method:After different concentrations allicin,oxaliplatin and their combinations effect on the human cerical cancer cells Hela,CCK assay was used to detect the growth inhibitory effect ,TUNEL and Annexin-V were used to evaluate apoptosis of Hela cells and the gene mRNA of bcl-2,bax and caspase-3 were examined by RT-PCR.Result:CCK showed that the growth of Hela cells was inhibited obviously by oxaliplatin in the range of 4~64μmol/L,and the inhibitory effect was depent on the time and the concentration,the IC50 of oxaliplatin on the time of 24h,48h and 72h was 62.4μmol/L,30.2μmol/L and 18.6μmol/L respectively, after the combination of allicin inhibition enhanced and IC50 of oxaliplatin on the time of 24h,48h and 72h were reduced to 40.6μmol/L,18.7μmol/L and 10.8μmol/L and the inhibition hasd enhanced 1.60,1.61 and 1.72 times respectively.Apoptosis cells were observed by FCM, The rate of apoptosis was increased from 22.7 +3.4% to 50.5 +5.7% after the combination of allicin (p <0.05), the rate of apoptosis were significantly different between experimental group and control group.TUNEL had the smilar result.RT-PCR tests showed that the mRNA of bcl-2 decreased while those of bax and caspase-3 increased after allicin and oxaliplatin .Conclusion::The growth of Hela cells was inhibited obviously by Oxaliplatin and was induced to apoptosis,and the inhibition and the rate of apoptosis increased when it conbinated with allicin,and its mechanism may be related to the downregulation of bcl-2 and the upregulation of bax and caspase-3.
