Objective: To investigate the expression and clinical significance of cell cycle regulator p21Waf1/Cip1 and S-phase kinase associated protein 2 (Skp2) in salivary gland tumors.Methods: The expression of p21Waf1/Cip1 and Skp2 in 10 cases of normal salivary gland tissues, 20 cases of benign salivary gland tumor tissues and 33 cases of malignant salivary gland tumor tissues were detected by immunohistochemical SABC method. Analyzing the relationship between their expression and clinicopathologic characteristics of malignant salivary gland tumor tissues, and their correlations to each other.Results: The expression rates of p21Waf1/Cip1 in normal salivary gland, benign salivary gland tumor and malignant salivary gland tumor tissues decreased gradually, and the differences were significant(χ2=11.113, p=0.006). The expression of Skp2 was not detected in normal salivary gland tissues, and the positive rate of Skp2 in the malignant salivary gland tumors(75.76%) was significantly higher than that in the benign salivary gland tumors (35.00%)(χ2=19.128, p=0.001). There was not relationship between p21 Waf1/Cip1 and clinicopathologic characteristics of malignant salivary gland tumors; but some relationship between the expression of Skp2 and lymph nodes metastasis(p<0.05). There was a inverse correlation between the expression of p21Waf1/Cip1and Skp2 in the benign salivary gland tumors, but the correlation was not significant(r=-0.390, p=0.089). There was a significantly inverse correlation between their expression in the malignant salivary gland tumors (r=-0.467,p=0.006).Conclusions:The low expression of p21Waf1/Cip1 and overexpression of Skp2 was highly associated with the occurrence and development of salivary gland tumors.Skp2 may take part in the development of salivary gland tumors through downregulating the expression of p21Waf1/Cip1.
