Expression Of Adioponectin Receptor 2 In Abdominal Subcutaneous Adipose Tissue And Placental Tissue Of Patients With Preeclampsia

Objective To investigate the role of adioponectin receptor 2(AdipoR2) in the pathogenesis of preeclampsia.Methods The expressions of AdipoR2 mRNA and its protein in abdominal subcutaneous adipose tissue and placental tissue in 20 normal pregnant women(control group) and 20 patients with preeclampsia(preeclampsia group) were measured by using real-time quantitative polymerized chain reaction (RT-qPCR) and Western blot analysis, respectively.Results We observed hyperinsulinemia and insulin resistance existed in preeclampsia group. AdipoR2 mRNA and its protein expressed in placental tissues and abdominal subcutaneous adipose tissues in both groups. As compared with control group,the levels of AdipoR2 mRNA and its protein in placental tissues were significantly higher in preeclampsia group (P<0.05). But the levels of AdipoR 2 mRNA and its protein in abdominal subcutaneous adipose tissues were significantly lower in preeclampsia group (P<0.05).The placenta AdipoR2 mRNA and its protein concentrations in both groups were no significantly correlated with prepregnancy body mass index(BMI), fasting blood glucose (FBG), homeostasis model assessment (HOMA) score, fasting insuline (FINS) (P>0.05). Though the levels of AdipoR 2 mRNA abdominal subcutaneous adipose tissues in normal pregnancy were no significantly correlated with pregnancy BMI, FINS, HOMA score(P>0.05), the levels of AdipoR 2 mRNA in abdominal subcutaneous adipose tissues in preeclampsia was inversely correlated with pregnancy BMI, FINS, HOMA score (P<0.05). No significant correlation was detected between FBG and AdipoR2 mRNA expressed in abdominal subcutaneous adipose tissues in preeclampsia (P>0.05).Conclusion AdipoR2 expression is up-regulated in placenta tissues but down-regulated in abdominal subcutaneous adipose tissues in preecalmpsia. These data suggest that adipoR2 plays a important role in mediating the effects of adiponectin in specific tissues in the pathogenesis of preeclampsia.Though AdipoR2 expression may not affected by weight,insulin ,insulin resistance in placenta tissues , but the level of AdipoR2 mRNA expressed in abdominal subcutaneous adipose tissues in preeclampsia may affected by overweight, hyperinsulinemia, hypoadiponectinemia, and insulin resistance.
