Objective: To investigate the expression of TLR4 in neonatal cord arterial blood mononuclear cells and TNF-αin the serum thus to explore the mechanism of TLR4 in preeclampsia.Methods: Twenty patients suffering from preeclampsia and twenty normal pregnancy patients were divided into two groups: experimental group and control group. Separating neonatal cord arterial blood mononuclear cells, the TLR4 mRNA and protein expression were measured by real-time quantitive PCR and Western Blot method, the expression of TNF-αin the serum was detected by ELISA.Results: The TLR4 mRNA level in neonatal cord arterial blood mononuclear cells (2-ΔCT:0.0669±0.0220), TLR4 protein expression (OD ratio: 0.81±0.16%) and TNF-αlevel in the serum(9.744 pg/ml±1.362 pg/ml)were all increased in experimental group compared with the control group, the differences were statistically significant (P<0.05). The expression of TLR4 mRNA and TNF-αin experimental group and control group showed positive correlation,the correlation coefficients were 0.52 and 0.53, respectively (P<0.05).Conclusion: The expression of TLR4 in neonatal cord arterial blood mononuclear cells and TNF-αin the serum were all increased in the experimental group. The expression of TLR4 and TNF-αin experimental group and control group showed positive correlation. TLR4-mediated release of inflammatory cytokines may be one of the important reasons leading to preeclampsia.
