Bronchial Asthma is a kind of commonly-seen clinical disease which frequently occurs in recent years, both of whose morbidity and mortality have an uptrend. It is one of the major diseases that has been seriously threatening people physically and mentally. Anti-asthma droppills which own anti-inflammatory, immunosuppressive pharmacological effects for asthma treatment consist of tripterygium wilfordii, sappan wood and borneol. The study utilizes the new method and means to systematically research on extraction, purifying the effective component of tripterygium wilfordii in anti-asthma droppills. In formula tripterygium extract is principal agent and acetoacetate extract of lignum sappan strangens the immunosuppressive pharmacological effects of tripterygium extract while borneol depresses the toxicity of both tripterygium extract and Lignum Sappan extract, enhancing curative effect.In this research, to extract effective constituent and remove impurity, the crude ethyl acetate extract of tripterygium wilfordii has been futured purified by neutro-alumina column chromatography, and the L9(3~4) orthogonal design was used to inspect the result of purification of tripterygium wilfordii extracting which was effected by the alcohol concentration, times, time and volumn and then, make sure the best craft of purification.Firstly in the preliminary experiment, droppills were made by solid dispersion with the bases of PEG6000 and F-68; and secondly to inspect the different condensing agents, different temperatures of physic liquor and different drop conditions for forming craft, through the experiment of mono-factor we obtained the final craft of anti-asthma droppills: dripper inside diameter/outside diameter is 3mm/4.5mm; condensing agent is dimethiconeⅠ; the temperature of physic liquor is 80℃; dropping distance is 4~5cm and dropping velocity is 36~45 drop/min; the length of condensing agent is 70cm; the face temperature of condensing agent is 18±2℃and the bottom temperature is 0±2℃; Thirdly the orthogonal design was used to screen the best formula (extract:PEG6000:F-68=1.5:4:1) when the index was degree of hardness , dissolution and degree of round. Fourthly, through the study of three batches samples, we improved the feasibility of the technology.On the base of optimized prescription, the output rate is high and it conforms to the standard of the Pharmacopoeia. In the study of the anti-asthma droppills quality evaluation, a thin layer chromatography method for qualitative distinguishing tripterygium wilfordii, borneol and sappan wood in the prescription medicine was developed, and the content of triptolide was determined by HPLC, the method has good specificity. We can control the quality of anti-asthma droppills effectively.The pharmacodynamics study showed that anti-asthma droppills had exact effect in the treatment of allergic asthma caused by histamine.In sum, the experiment will provide the scientific foundation of the big industries'production, lay a good foundation for the researchment and exploration of new medieine, and offer the reference of stnadardization of herbs.
