Fructus Arctii is the dried ripe fruit of Arctium Lappa Linne (Compositae) and is commonly used.Through antiviral screening in vitro of different part of Fructus Arctii, we found the active part is total lignanoids. Through optimum choice on different extracting methods by orthogonal experimental design and homogeneous experimental design, we got the optimum extraction technology and the total lignanoids in Fructus Arctii extract could amount to more than 80 percent.Determining on the arctiin content of its extract through HPLC and the total lignanoids content through ultraviolet spectrometry, the same time, carrying on methods investigation to found the standard quality control of Fructus Arctii extract.We studied on antiviral action of Fructus Arctii extract through establishing virus model by vaccinating influenza virus adaptive strain (A/FM/1/47) on mouse's lung. Results shows that Fructus Arctii extract could remarkably decrease the infectivity of influenza virus after 48thours and the restrain action of Fructus Arctii extract on influenza depends on its concentration through prevention administration, the optimum concentration is 0.0125 g/ml .It also indicates that if mixing the virus with drug solution, each concentration under the maximum toxic dose has strong restrain action on influenza virus.We also studied on substance base in antiviral extract, which are chemical compositions. We got four compounds from it and identified the structures according to physical-chemical properties and spectrum data. They are arctigenin,, Lappaol F, Arctignan E and arctiin. Carrying on purity determination through HPLC and confirming each composition's location on HPLC spectrum of the extract.As there are two kinds of commercial material medicine of Fructus Arctii in market: crude and prepared drugs, the content of arctiin and total lignanoids may be different. In the paper, studies on different preparation methods of Fructus Arctii and comparing the changes of content of total lignanoids, arctiin and arctigenin .We also made comparasion studies of IR spectrum and HPLC spectrum on the raw and prepared materials, which has laid a good foundation for the normalization of preparation methods of Fructus Arctii.This article will also lay a foundation for the development of Fructus Arctii on the national antivirus new drug of V.
