Biomembrane chromatography, which uses cell membrane of human or animal as a stationary phase, is a good technology for studying the interactions between drug and cell membrane, membrane receptor and enzyme. In this way, it retains best the integrity and enzymatic activity of cell membrane, the stereo-structure and surroundings of membrane receptor. The characteristic of drug-receptor interactions can be determined from chromatographic date. The cell membrane chromatography needs no labeled a highly radioactive element and the membrane receptor is presented in nature state. The preparation is more convenient than artificial membrane chromatography and the column could be reused over a long time.However, it is very strict to the preparation of stationary phase and the working conditions such as mobile phase, pH, and column temperature. In our present work, a novel capillary biochromatography system via sol-gel technology was developed, and the biomembrane-drug interactions of different biomembranes were studied. There are three chapters in this thesis.In chapter 1,the development of the capillary electrophoresis was introduced, and the sol-gel technology, the application of biochromatography was also described.In chapter 2, the investigation of biomembrane-drug interactions of different biomembranes by capillary electrophoresis was carried out. Effects of immobilized biomembrane amount, pH and biomembrane species including mice red cell membrane, human red cell membrane and S-80 cell membrane on the interactions between drugs and biomembranes had been investigated. The availability of this system to the study of biomembrane-drugs interactions and screening of new drugs was evaluated.In chapter 3,the separation ofβ-lactam antibiotics by capillary biomembrane chromatography was carried out and the optimal condition for separating was established.The results showed that the chromatography system might be used for simulating the interactions between drugs and the membrane in vivo.It might be a very useful tool for tracking the action mechanism of drugs and screening of new drugs.
