Objective:Interleukin -17 (interleukin-17, IL-17) is a multifunctional cytokine, which has a strong recruitment of neutrophils to promote the release of inflammatory cytokines in a variety of cells and other biological effects. Interleukin -8 (interleukin-8, IL-8) have increased in many inflammatory diseases, especially in most of the interstitial proliferation. To detect the expression of IL-17and IL-8 in simple benign prostatic hyperplasia(BPH) and BPH combined with prostatitis,and to evaluate the effect of inflammation on the development and progressionof BPH.Materials and methods:1. Obtained specimens of 60 patients with BPH from the First Affiliated Hospital of Fujian Medical University after underwent transurethral resection of prostate (TURP).2. 60 specimens were divided histologically by HE staining into group of simple benign prostatic hyperplasia ( A group) and group of benign prostatic hyperplasia combined with prostatitis(B group)3. Protein levels expression of IL-17 and IL-8 were examined by immunohistochemical.4. mRNA levels expression of IL-17 and IL-8 were examined by Using Real-time fluorescence quantitative PCR (RT-PCR).Results: (1) immunohistochemical staining: IL-17 and IL-8 expressed in the A group and B group, but in the B group of tissues were significantly higher than A group of tissues (P <0.05); IL-17 and IL-8 expression in the B group of organizations had a positive correlation (R = 0.707 P <0.05) (2) IL-17mRNA in the A group, the relative ratio: 0.042±0.019,in the B group, the relative ratio: 0.108± 0.025;IL-8mRNA in the A group relative ratio: 0.303±0.076, in the B group, the relative ratio: 0.536±0.059, The expressions of IL-17mRNA andIL-8 mRNA in the B group were significantly higher than the expressions of A group (P <0.05) . Conclusion: Pro-inflammatory cytokines IL-17 and inflammatory cytokines IL-8 in the tissues of benign prostatic hyperplasia combined with prostatitis are significantly increased and the expression levels of both cytokines have positive correlation.
