BackgroundThe glucose-regulated proteins (GRP) refer to a set of proteins generated by cell in order to adapt to endoplasmic reticulum stress.GRP78 and GRP94 are important members of the tribe,whose main physiological function is involved in protein synthesis when the newborn polypeptide chain folding, as molecular chaperones.Recent studies show that, high expression of GRP was found in some tumor cells,they were involved closely in the generation, progression and prognosis of malignant tumors,and supply new theory support for clinical diagnosis and treatment for malignant tumors.ObjectiveTo study the expressions of GRP78 and GRP94 in rectal adenocarcinoma and the relationship between them and the clinical and pathological features of rectal adenocarcinoma,and its clinical significance.MethodsWe Collected 45 cases of surgically resected specimens in rectal adenocarcinoma and their pericancerous tissues for contrast. Specimens were confirmed by pathology exam and clinical data integrity.All the specimens were fixed in 10% formalin and routinely embadded in paraffin.EnVisionTM was used for assaying the expressions of GRP78 and GRP94,and scored them under high-power microscopy.Put the data into SPSS 13.0 and analyze the correlation between the expression level of GRP78 and GRP94 and extent of differentiation,clinical stage,depth of invasion and lymph node metastasis of rectal adenocarcinoma. Level of significance was set to 0.05. Results1.In 45 cases of rectal adenocarcinoma,positive expression of GRP78 and GRP94 were both 24 cases (53.3%); In 45 cases of pericancerous tissues positive expression of GRP78 and GRP94 were 6 cases (13.3%) and 7 patients (15.6%);In rectal adenocarcinoma GRP78 and GRP94 expression was significantly higher than pericancerous tissues (χ2= 16.200, P<0.01;χ2=14.221, P<0.01).2.The levels of GRP78 and GRP94 expression were correlated with extent of differentiation,clinical stage,depth of invasion and lymph node metastasis of caner(P< 0.05 or P<0.01),but not with patient's sex,age and size of tumor(P>0.05).3.In 24 cases that GRP78 expression's positive,the number of cases GRP94 expression's positive was 18;In 21 cases that GRP78 expression's negative,the number of cases GRP94 expression's positive was 6;The consistence of GRP78 and GRP94's expression was found.(r= 0.464,P<0.01).Conclusions1.GRP78 and GRP94 were overexpressed in rectal adenocarcinoma.2.GRP78 and GRP94 maybe be involved in the generation, progression and prognosis of rectal adenocarcinoma.
