Objective: To investigate relationship between chemosensitivity assay in vitro and the expression of glucose- regulated protein 78(GRP78) and 94(GRP94) in non-small cell lung cancer (NSCLC), supplied the basis of chemotherapeutic drugs therapy in clinical.Methods: methylthiazo-letrazolium (MTT) assay were used to detect the sensitivity of 9 common-used chemotherapy drugs in 41 samples of NSCLC in vitro, and cell growth inhibiting rate and resistance rate were calculated. The expression of GRP78 and GRP94 were detected by immunohistochemistry method. Then, Spearman correlation assay was used to investigate the correlation between the cell growth inhibiting rate and expression of GRP78, GRP94.Results: The resistant rates of 9 drugs were as follows: PTX,VP-16,CBP,TPT,VCR,DDP,ADM,NVB and CTX resistance rate : 46.34%,56.09%,60.97%,60.97%,65.85%,78.04%,80.48%,87.80%,100.00%, respectively. 12 among 41 cases of NSCLC indicated the complete resistance to 9 chemotherapy drugs. There were no significant differences in different pathological type, differentiation and clinical stage on complete resistance.Immunohistochemistry results: There was a significant elevation of expression of GPR78 and GRP94 in NSCLC tissues compared to normal lung tissues. A stronger stain was observed in cancer tissue (main intensity of staining ++ - +++) compared to normal tissues (main intensity of staining + - ++).
