| Objective To investigate the expression of protein and mRNA of glucose-regulated protein 78 (GRP78) and glucose-regulated protein 94 (GRP94) in human gastric carcinoma and paracancer tissue at the cellular and molecular level, to analyze clinical pathological features relationship with them, explore their relationship with the biological behavior of gastric carcinoma.Methods (1)The expression of GRP78 and GRP94 proteins of gastric carcinoma (GRP78: 156 cases, GRP94: 149 cases) and 40 cases of adjacent normal tissues were detected by immunohistochemistry (s-p method) on TMAs (tissue microarrays). (2)Reverse transcription-polymerse chain reaction (RT-PCR) was applied to detect the expression of GRP78 mRNA and GRP94 mRNA of 30 cases of gastric carcinoma and 10 cases of paracancer tissue. (3) Both results of immunohistochemistry and RT-PCR were combined to clinical and pathological data to analyze the relationship between expression of them and development, invasion or metastasis of gastric carcinoma.Results (1) In gastric carcinoma and adjacent normal tissue, the positive expression rate of GRP78 protein were 80.1% (125/156) and 32.5% (13/40) respectively, there was significant difference between the two groups (P <0.05). The expressions of GRP78 protein were closely correlated with tumor size, depth of invasion, metastasis of lymph nodes and TNM stage (P <0.05), but no association was observed with the gender, age, and pathological grades. The positive rate of GRP78 mRNA in gastric carcinoma was significantly increased relative to adjacent normal tissue (P <0.05).(2)In gastric carcinoma and paracancer tissues,the positive expression rate of GRP94 protein was 73.8%(110/149)and 30.0%(12/40) respectively, there was significant difference between the two groups(P <0.05). The expressions of GRP94 protein were closely correlated with tumor size, depth of invasion, metastasis of lymph nodes and TNM stage(P <0.05), but no association was observed with the gender, age, and pathological grades. the positive rate of GRP78 mRNA in gastric carcinoma was significant higher than adjacent normal tissue(P <0.05). The expressions of GRP94 mRNA was significantly related with pathological grades (P <0.05).Conclusion (1) Protein of GRP78 and GRP94 both stablely express in gastric carcinoma. They were closely related with the development of gastric carcinoma, and can serve as useful indicators to determine the malignant of gastric carcinoma.(2) Both the mRNA of GRP78 and GRP94 are highly expressed in gastric cancer tissue, but they may have relationship with the regulation of transcription. |
