Purpose:To investigate the relationships between the expression of glucose-regulated protein 94 and 78(GRP94, GRP78) in human NSCLC;and the correlations with clinical data.Object:Lung cancer tissues and paracancer normal lung tissues, were collected from 64 patients undergoing surgical resections of lung cancer at the China-Japan friendship hospital of Ji Lin University. The Samples include 43 SCC pathological sections, 21 ACC pathological sections.Methods:Telephone following up regarding on life span and postoperative treatment was carried out with 64 patients who visited at China-Japan friendship hospital of Ji Lin University. Immunohisto- chemisty was used on 64 NSCLC pathological sections to detect the expression of GRP94 and GRP78.Results:1.pathology diagosis of cell differentiations may influence the survival rate on pulmonary adenocarcinoma;tumor T /N staging and WHO clinical stage are the most factors closely correlated with the survival rate in both lung squamous cell carcinoma and adenocarcinoma.2.There were significant overexpressions of GRP94 and GRP78 at protein levels in lung cancer tissues as compared to adjuncent normal tissues in NSCLC;the difference for GRP94 and GRP78 protein expression between cancer tissues and paracancer tissues was significant.The protein expression of GRP94 and GRP78 correlated with pathology differentiation,tumor T staging,tumor N staging,WHO clinical staging.3.The overexpression of GRP94 and GRP78 in NSCLC correlated with life span and survival rate, which showed that the higher expression of GRP94/78, the poorer survival rate,which suggested that cell ER stress factors GRP94 and GRP78 can be used as an indicator for prediction of prognosis.4. The GRP94 and GRP78 were overexpressed in more than 30% of tested cancer tissues,which suggested above lung cancer correlators act some roles in NSCLC;GRP94 and GRP78 proteins were overexpressed in more than 50% of tested paracancer tissues, which suggested they involed in neoplasm invasiveness.5.The difference for GRP94 and GRP78 protein expression between cancer tissues and paracancer tissues were significant in NSCLC,and there were relationships among the protein expression of GRP94 and GRP78.6.In NSCLC,the protein expression of GRP94 and GRP78 correlated with pathology type, pathology differentiation, TNM clinical staging,tumor N staging,tumor T staging,smoking history,Ki67 expression.Conclusions:GRP94 and GRP78 might be a potential indicator for survival of NSCLC. The expression level of GRP94 may can be served as an additional markers for identifying the grade of differentiation, while GRP78 may be used for indicating the clinical stage on diagonosis of NSCLC.
