Objective:The purpose of this in vitro study was to investigate the effect of arecoline on the expression of uPARAP in human oral mucosal fibroblasts, further to explain the effect of arecoline on the pathogenesis of OSF.Method:1. The fibroblasts were obtained from normal human oral mucosa tissues and cultured in vitro. Cells of third to tenth passages were selected for using in the following experiment. According to the concentration of arecoline in cell culture medium, the cells were randomly divided into five groups, Oμg/ml,5μg/ml,10μg/ml, 25μg/ml, and 50μg/ml.2. MTT assay was used to detect the proliferation of fibroblast in five different groups after cultrued for 12,24,48,72 hours.3. Treated with arecoline at different concentration for 24hours, the expressions of uPARAP were observed by immunofluorescence.4. Treated with arecoline at different concentration for 24hours, the percent of expression of uPARAP were observed by flow cytometry. Results:1. Compared with control group 0μg/ml, cells in Group 5μg/ml and 10μg/ml have no difference in proliferative potentiality; while cells in Group 25μg/ml and 50μg/ml show dificency in proliferation, especially Group 50μg/ml.2. The uPARAP is expresssed lower in 5μg/ml group, 10μg/ml group, 25μg/ml group and 50μg/ml group after treated with arecoline at different concentration for 24hours detected by immunofluorescence and flow cytometry. The reduction is more significant in 25μg/ml group and 50μg/ml group, especially in 50μg/ml group.Conclusion:1. Arecoline can affect the proliferation of oral mucosa fibroblasts.2. High concentrations of arecoline significantly can inhibit the expression of fibroblast uPARAP.
