Experimental Study Of Jianpi Yizhi Method On The Expression Of Aβ、GFAP In Hippocampal Region Of The Rat With Synthetic Alzheimer Disease

Objective:To observe the effect of Jianpi Yizhi Capsule on ethology, the serum blood acetylcholin esterase(AChE), the number of Aβ1-40 positive cell and on expression of GFAP of Astrocytes in hippocampal region of the rats with synthetic Alzheimer disease, and explore the therapeutical effect on AD rats and the Potential mechanism of action of Jianpi Yizhi Capsule.Methods:Fifty-two sensitive SD (sPragu-dawleg) rats were classified into five Groups: the blank control group, sham- operated group, model group, Chinese herbal medicine group (CHM),western medicine group(WM).The Alzheimer disease model was established by Aβ1-40 injected into hippocamal in rats.The faculty of leaning and memory was evaluated by shuttle-box test and Morris water maze experiment after establishing AD model four weeks and administer drugs four weeks. ELISA method to measure the serum blood acetylcholin esterase(AchE),The brain tissues were removed to be made paraffin wax sections,the number of Aβ1-40 positive cell and on expression of GFAP of Astrocytes in hippocampal region was measured with immunohistochemical staining.Results:(1)After establishing AD model four weeks,the results of shuttle-box test. showed that compared with the blank control group, the number of electric shocks and passive avoidance times of model group,CHM, WM is significantly increased(P<0.01).Morris water maze experiment, the hidden platform of model group,CHM, WM was longer than the blank control group (P<0.01), the average pass through times significantly decreased (P<0.01). (2)After administer drugs four weeks,the results of shuttle-box test showed that compared with the model group, the number of electric shocks of CHM, WM’ times to evade stimulate is least(P<0.01).Morris water maze experiment, the hidden platform of CHM, WM was shorter than the model group (P<0.01), the average pass through times significantly increased (P<0.01). (3) The results of measure the serum blood AchE:Horizontal comparison, the contents of AChE of model group,CHM, WM have significant differences compared with the blank control group (P<0.01)after establishing AD model four weeks. No significant difference was found in the CHM, WM comparison with the blank control group(p>0.05).Significant difference was found in the CHM, WM comparison with the model group(P<0.01) after administer drugs four weeks.Transverse comparison, there is Significant difference between before administer drugs and after(P<0.01).(4)The results of Aβ1-40 immunohistochemistry indicate:the number of Aβ1-40 positive cell of CHM, WM comparison with sham-operated group have no significant difference (P>0.05).And significantly decreased in comparison with the model group(P<0.05). (5)The results of GFAP immunohistochemistry indicate:the number of GFAP positive cell of CHM, WM comparison with the model group significantly increased and have significant difference (P<0.05).Conclutions:(1)The Presentation of results of the study that the model was established by Aβ1-40 injected into hippocamal was blest, because the model rats had the changes of Praxiology and Pathology. (2)Ethology indicated that Jianpi Yizhi Capsule can improve the learning and memory of the model rats.(3)Jianpi Yizhi Capsule can suppress the activity of AchE,increased the contents of Ach in rats. Protect cholinergic neurons, improve the learning and memory abilities.(4) Jianpi Yizhi Capsule can significantly decreased the number of positive cell. An efficient decreased to Aβ’s deposit. (5) Jianpi Yizhi Capsule can improve the expression of GFAP in hippocampal region, promote activation of Astrocytes in the AD rats.Those partly may be the therapeutic mechanism on Alzheimer disease of Jianpi Yizhi Capsule.
