Purpose:To evaluate the expression pattern of SHIP2 protein in tissue specimens and cell lines of laryngeal squamous cell carcinoma(LSCC), and further correlate SHIP2 protein expression with clinicopathological parameters and prognosis in LSCC.Methods:Western blot was applied to assess SHIP2 protein expression in the LSCC cell line Hep-2 and the nontransformed nasopharyngeal epithelial cell line NP-69. Immunohistochemical staining was performed on paraffin-embedded tissue specimens to investigate SHIP2 protein expression, which included 54 cases of LSCC specimens and 16 cases of adjcent tissue samples.Results:A significant increased SHIP2 protein expression was identified in both LSCC tissue and cell levels and its overexpresion was tightly associated with LSCC T classification (x2=10.119, P=0.006), clinical stage (Ⅰ+Ⅱ/Ⅲ+Ⅳ) (x2=13.460, P=0.001), metastasis (x 2=12.959, P=0.002) and recurrence(x2=11.241, P=0.004). Patients with SHIP2 overexpression had both poorer disease-free survival and overall survival than that in patients with low SHIP2 expression(median survival, 41.231±3.164 months versus 56.036±2.267 months, respectively; P<0.001). Multivariate Cox regression analysis revealed that both SHIP2 expression and lymph node metastasis in LSCC were independent prognostic factors for patients with LSCC.Conclusions:This is the first report to demonstrate that SHIP2 protein expression was obviously increased in LSCC tissue and cell levels, and SHIP2 protein overexpression was associated with tumor recurrence, metastasis and poorer prognosis in LSCC patients. These results revealed that SHIP2 might play a critical role in the initiation and progression of LSCC, implicating SHIP2 as a valuable marker for prediction in LSCC.
