The invading and metastasis of tumour plays important role when the tumour happens and develops. Definite evidence indicates that more than 90% of the cancer patient dies of tumour metastasis or the relapse ultimately. In metastasis process, DKK-1 is playing an important role. DKK-1 is able to regulate and control the signal transmiting by combining with its corresponding acceptor. DKK-1 can also induce the apoptosis of tumor cells by the classical signal of Wnt/β-chain of rings protein (beta-catenin) conduction approach as well as Wnt which is not the classical signal conduction approach within the body. DKK-1 can also regulate and controll the various tumour cell from bone metastasis as well as facilitate the infiltration and metastasis of some tumour cells , which plays very important effect in the field of tumour occurrence and development. Ovary cancer is one of the most common cancer in department of gynaecology ,whose death rate takes up of department of gynaecology tumour . The ovary tumour is extremely easy to metastasis and irradiate. While 90% of the ovary cancer originates from epithelium ,whose early phase is not easy to be found and extremely easy to metastasis. Therefore, we have fostered the ovary cancer cells to be HO-8910 and HO-8910-PM and collected the epithelium tumour specimen from Xi Jing hospital operation in 2007- 2009 .Our aim is study the different expression for DKK-1 in different ovary cancer organization and study the expressing of DKK-1 in ovary cancer .We also have studied it's relation with the differentiation degree,infiltration and lymph node in ovary tumour .Futhermore , we deal the ovary cancer cells HO-8910-PM with adriamycin .Then we study the different expression for DKK-1 in these cells and the change for the invasion of the cancer,which may provide a basis to the method in studying the restraint tumour changes and invades further.AIM:1. Foster the ovary cancer cell to be HO-8910 and HO-8910-PM , and study the expression of DKK-1 protein in it's surface;2. Collect 80 regular operation specimen for ovary epithelium tumours , then study the expression of DKK-1 protein in it's surface.3. Study the expression of DKK-1 in the cells HO-8910-PM which was dealed with adriamycin,then study the change for the invasion of the cancer.METHODS:1.Immunocyte chemistry,Western blot law and RT-PCR method are used to detect the expression of DKK-1 protein in ovary cancer cells;2. SABC immunohistochemical method and RT-PCR method was used to detect the expression of DKK-1 protein in human ovarian tumor .3.Immunocyte chemistry method is used to detect the expression of DKK-1 protein in ovary cancer cells HO-8910-PM which was dealed with adriamycin.Transwell method is used to study the change for the invasion of the cancer.RESULTS:1. The DKK-1 protein expresses higher in HO-8910 ovary cancer cell than in the HO-8910-PM ovary cancer cell;2. DKK-1 protein expresses higher in the ovary cancer organization with lymph node changes than which without the lymph node changes.3. DKK-1 protein expresses higher in the ovary cancer cells HO-8910-PM which was dealed with adriamycin,meanwhile,the ablilty for the invasion of the cancer is decreased.CONCLUSION:1. DKK-1 protein is expressed in ovary cancer cells as well as ovary cancer organization;2. The low expression of DKK-1 protein is related to the invasion and lymph node metastasis in ovary cancer.
