Abstract:bjective:Through experiment to explore the theory of " Shaoyang in charge of the bone" in Huang di's Internal Classic,which has been buried for long time. That were two different theory with " the kidney in charge of the bone " and " Shaoyang in charge of the bone ". Observing the effecs of the Foot-Shaoyang Channel-Points can modulate bone mineral density(BMD) and bone metabolism markers with osteoporosis rats. With the change of bone remodeling and metabolism,clarify the role of modulation bony strength and improvement osteoporosis. Initial studying the path and mechaism with prevent and treat osteoporosis.Methods:40 female Wistar rats aged 12 months, weight (220±10g).The room temperature was kept in 25±1℃.This rats were randomly divided into 4 groups:Sham group,OVX Control group(OVX-C), OVX Acupuncture group(OVX-A) and OVX Non-point Acupuncture group (OVX-N),10 in eaeh group. The Sham group was only sham operated and the other four were ovariectomy.After the operation,We have feed the four groups rats daily. Both OVX-A and OVX-N were conducted treatments three months After the operations.We applied Yanglingquan,Huantiao,Xuanzhong,Jingmen for the OVX-A group;Non-points with the interior posterior limb for the OVX-N group,There wasn't any therapy in Sham group and OVX-C group; Giving Electroneedling therapy once a day. Three months later after treating, take every group rats,total RNA was isolated from left femurs. Expressions of OPG,RANKL and CBFA1mRNA were detected by semiquantitative RT-PCR. Dual-energy X-rays (DEXA) were used to test the BMD of right femurs. Blood samples were taked from the hearts and Enzyme-linked Immunoadsordent Assay (ELISA) method to test the level of NTX,PINP,BGP. Urine were tested the level of DPD. Results:1.Three months later after treating,the level of BMD/body weight adjust value were decreased in the OVX-N and OVX-C, with significant difference as compared with that in OVX-A and Sham group (P<0.01),and no significant differences between OVX-A and Sham group(P> 0.05).2.Three months later after treating,the relative amount of OPGmRNA expression were decreased in the OVX-N and OVX-C, with significant difference as compared with that in OVX-A and Sham group (P<0.05),and no significant differences between OVX-A and Sham group(P>0.05).3.Three months later after treating,The relative amount of RANKLmRNA expression were increased in the OVX-C, with significant difference as compared with that in Sham group(P<0.05).The relative amount of RANKLmRNA expression were decreased in the OVX-A group,with significant difference as compared with that in the OVX-N and OVX-C (P<0.05), no significant differences with Sham group(P>0.05).4.Three months later after treating,the relative amount of OPG/RANKLmRNA expression were decreased in the OVX-N and OVX-C group,with significant difference as compared with that in OVX-A and Sham group (P<0.01),and no significant differences between OVX-A and Sham group(P>0.05).5.Three months later after treating,the relative amount of CBFA1mRNA expression were decreased in the OVX-A and OVX-N, with significant difference as compared with that in Sham group (P<0.05),and the relative amount of CBFA1mRNA expression were decreased in the OVX-C, with significant difference as compared with that in Sham group (P<0.01).6.Three months later after treating,the level of DPD were increased in OVX-C group,with significant difference as compared with that in OVX-A,OVX-N and Sham group (P<0.05);and there no significant differences in OVX-A,OVX-N and Sham group(P>0.05).7.Three months later after treating,there were no significant differences between groups in the level of NTX.8.Three months later after treating,the level of PINP were decreased in OVX-C and OVX-N,with significant difference as compared with that in Sham group (P<0.05);and no significant differences between OVX-A and Sham group(P>0.05).9.Three months later after treating,there were no significant differences between groups in the level of BGP. Conclusion:Foot Shaoyang Channel-point with electroneedling can surely have antiosteoporotic effect.
