Objective: To observe insulin-resistant rats adiponectin levels and LPtin levels,liver adiponectin receptor expression and liver LPtin receptor expression,and study the Dahuang Zhechong Pill on the prevention and treatment of insulin resistance and its mechanism.Insulin resistance is a common risk factor for metabolic syndrome which includes a series of pathological states of obesity,diabetes,hypertension,atherosclerosis,lipid metabolic disorders The fat cells is not only the energy reserves organ, but also secrete various cytokines endocrine organ.It plays an important role on metabolism and energy balance.Most of adipocytokines were an important factor leading to IR and risk factors in cardiovascular Disease.Some cytokines can prevent the occurrence of IR,while others can promote the formation of IR. The former has adiponectin,the latter includes LPtin, TNF-α, resistin.Dahuang Zhechong Pill comes from a work by Zhang Zhongjing. With the study of medicine,this research used fat-rich feeding rats to reproduce IR model and explore the effect of Dahuang Zhechong Pill on IR and mechanisms of improve IR,Providing a theoretical basis for clinical research and development of new drugs to improve the condition of IR.Method: 50 rats were divided into 5 groups randomly :control group(A group), model group (B group), low-dose group(C group), high-dose group (D group)and Xue Zhi Kang group(E group).The rats in the control group were fed with normal forage. Others were fed with forage of high cholesterol in control group and model group were fed with 10ml distilled water per Kg The rats in high-dose group were fed with 2g Dahuang Zhechong Pill per 1kg body weight, and in low-dose group with 1g.Xue Zhi Kang group:Xue Zhi Kang capsule 0.2g/kg.All animals in five groups were fed 8 weeks continuously.At the end of the 8th week,blood was collected for being measured biochemical markers and serum adiponectin and LPtin detection were measured.Prompt the liver cryopreserved to be used the measure of ADIPOR2mRNA,OB-R mRNAResults:1 GLU,INIS,HOMA-IR,ISICompared with control group,Glu,INS,HOMA-IR increased(p<0.01), while ISI decreased obviously in model group(p<0.05); Compared with model group,Glu,INS,HOMA-IR decreased significantly(p<0.01),while ISI was increased significantly in low-dose group and high-dose group(P<0.01); Compared with low-dose group,Glu,INS,HOMA-IR decreased,while ISI was increased in high-dose group,better than low-dose group,but there were no statistical significance(P>0.05); In low dose-group and high-dose group compared with Xue Zhi Kang group,high-dose group Glu decreased significantly(p<0.05),HOMA-IR decreased,and ISI was increased,better than Xue Zhi Kang group,but there were no statistical significance(P>0.05).2 TC,TG,LDL-C,HDL-CCompared with control group,TG,TC,LDL-C increased(p<0.05),while HDL-C decreased obviously in model group(p<0.01); Compared with model group,TG,TC,LDL-C decreased significantly(p<0.05),while HDL-C was increased significantly in low-dose group and high-dose group(P<0.01); Compared with low-dose group,TG,TC,LDL-C decreased in high-dose group, better than low-dose group,but there were no statistical significance(P>0.05),while low dose group HDL-C was increased,better than high -dose group,but there were no statistical significance(P>0.05); In low-dose group and high-dose group compared with Xue Zhi Kang group,TC,TG,LDL-C increased,better than Xue Zhi Kang group, but there were no statistical significance(P>0.05).3 The serum adiponectinCompared with control group,APN decreased obviously in model group(p<0.01); In low-dose group and high-dose group compared with model group, In high dose group APN increased significantly(p<0.05),while low-dose group APN was increased,better than model group,but there were no statistical significance(P>0.05); In high-dose group compared with low-dose group, APN increased,better than low-dose group,but there were no statistical significance(P>0.05); In low-dose group and high-dose group compared with Xue Zhi Kang group,In high-dose group APN increased,better than Xue Zhi Kang group,but there were no statistical significance(P>0.05).4 The serum LeptinCompared with control group,LP increased obviously in model group(p<0.01); In low-dose group and high-dose group compared with model group, LP decreased obviously(p<0.01); In high-dose group compared with low-dose group, LP decreased,better than high-dose group,but there were no statistical significance(P>0.05); In low-dose group and high-dose group compared with Xue Zhi Kang group, LP decreased,better than Xue Zhi Kang group,but there were no statistical significance(P>0.05).5 The mRNA expression of AdipoR2 in liver muscleCompared with control group, AdipoR2 mRNA expression decreased in model group(p<0.01); In low-dose group and high-dose group compared with model group,high-dose group AdipoR2mRNA expression increased significantly(p<0.01),low-dose group AdipoR2 mRNA expression increased significantly(p<0.05); In high-dose group,compared with low-dose group, AdipoR2 mRNA expression increased,better than low-dose group,but there were no statistical significance(P>0.05); In low-dose group and high-dose group compared with Xue Zhi Kang group,high-dose group AdipoR2 mRNA expression increased,better than Xue Zhi Kang group, but there were no statistical significance(P>0.05).6 The mRNA expression of OB-R in liver muscleCompared with control group, OB-R mRNA expression decreased in model group(p<0.01); In low-dose group and high-dose group compared with model group,high-dose group OB-R mRNA expression increased significantly(p<0.01), while low-dose group OB-R mRNA expression increased,better than model group,but there were no statistical significance(P>0.05); In high-dose group,compared with low-dose group, OB-R mRNA expression increased,better than low-dose group,but there were no statistical significance(P>0.05); In low-dose group and high-dose group compared with Xue Zhi Kang group,high-dose group OB-R mRNA expression increased,better than Xue Zhi Kang group, but there were no statistical significance(P>0.05).Conclusion: 1 High fat diet can induce rat model of insulin resistance, indicating the existence of IR are closely related to lipid abnormalities.2 Dahuang Zhechong Pill can improve insulin sensitivity in IR rats, control IR.3 Dahuang Zhechong Pill can increase IR rats serum Adiponectin levels.4 Dahuang Zhechong Pill can decrease IR rats serum Leptin levels.5 Dahuang Zhechong Pill can increase the mRNA expression of Adiponectin receptor in the liver of IR rats.6 Dahuang Zhechong Pill can increase the mRNA expression of Leptin receptor in the liver of IR rats.
