1 ObjectiveObserved the effection of Touguxiaotong Capsule(TXC) on histomorphology of subchondral bone in rabbit knee osteoarthritis, and to explore the mechanism of TXC on the role of osteoarthritis(OA).2 Methods2.1 Grouping and treatment with medicine:48 healthy New Zealand rabbits aged 6 months, which were randomly divided into 4 groups, i.e.,the normal group (12 rabbits),the model group (12 rabbits),the control group (12 rabbits) and the experimental group (12 rabbits).Except for the normal group,the other three groups were treated in one week after operation.The rabbits belonged to the normal group had no treatment. The model group,the control group and the experimental group were respectively perfused with normal saline 10ml·d-1, the water-solution of Zhuangguguanjiewan 10ml·d-1(contained medcine 1.12g)and TXC 10ml·d-1 (contained medcine 1.12g).After operation,each rabbit was sacrificed respectively in 6,9 and 12 weeks.2.2 Observational indexes:Through the general macroscopic observation and X-ray radiography of macroscopic observation, and observed subchondral bone and cartilage pathology of microscopic changes.3 Results3.1 Macroscopic observation:Articular cartilage appear blue and white in the experimental group,histo-pathological and synovial membrane inflammation is lower than the model group and the control group.3.2 X-ray and light microscopic observation:3.2.1 Comparison between the model group and the normal groupBy the end of the sixth week after operation, the medial knee joint space was narrow, osteophyte formation was not obvious in model group;Observation through light microscope indicated that the development of osteoporosis in subchondral bone of the model group. By the end of the ninth week after operation, medial gap narrowed significantly, obviously osteophyte formation, joint surface roughness deformation and hardening;histomorphology observation showed that disorder of bone architecture,micro-fracture of bone trabecula, osteoporosis and bony repair occurred in subchondral bone. By the end of the twelfth week after operation, osteophyte formation was obvious,histomorphology observation showed the eburnation and redundancy in subchondral bone,the disorder of bone architecture and the serious cartilage degeneration.3.2.2 Comparison between the experimental group and the model groupBy the end of the sixth week after operation, the medial knee joint was narrow in experimental group, joint surface roughness deformation, joint edge of a slight osteophyte formation, subchondral bone density increased;observation through light microscope indicated that osteoporosis in subchondral bone in the experimental group were less serious than those in the model group. By the end of the ninth and twelfth week after operation,histomorphology observation showed slight osteoporosis in subchondral bone and slight cartilage degeneration, with occasional micro-fracture of bone trabecula in the experimental group in the ninth week. There were less serious eburnation in subchondral bone, better bone architecture and slight cartilage degeneration in the twelfth week.3.2.3 Comparison between the control group and the model groupBy the end of the sixth week after operation, the medial knee joint space was narrow in control group,ranging from model group and the experimental group;some of the denudation of articular cartilage, and cartilage thickness varies subchondral bone osteoporosis, trabecular bone fracture can be seen .By the end of the ninth and the twelfth week after operation, observation on subchondral bone through light microscope showed micro-fracture of bone trabecula and bone sclerosis were slighter in the control group in the ninth week after operation, so were bone sclerosis and cartilage degeneration slighter in the twelfth week.3.2.4 Comparison between the experimental group and the control groupBy the end of the sixth week after operation, the experimental group had a slight edge joint osteophyte formation, subchondral bone density increased. Observation through light microscope indicated that osteoporosis in subchondral bone in the experimental group were less serious than those in the control group. By the end of the ninth and the twelfth week, the results of histomorphology observation on the experimental group was better.4 According to the standards of articular cartilage Colanbo integral statistics show that the chondrocytes in the experimental group was rated better than the same period in the model group, control group was significantly lower in 9,12 week and there was significant statistical difference (P<0.05).4 Conclusion4.1 There is always pathological changes in the subchondral bone during the process of the occurrence,and development of osteoarthritis.We initially proof that the evident strengthening of changes of subchodral bone in OA is a process from bone-resorption-oriented in prophase and bone-formation-oriented in anaphase,to the final subchondral bone sclerosis.4.2 TXC to reduce cartilage and subchondral bone damage, slow down the development process of OA,may be reduced by a certain part of subchondral bone to reduce the incidence of early osteoarthritis subchondral bone resorption, inhibit the latter part of the pathogenesis of osteoarthritis subchondral bone formation, thus improving the subchondral bone morphogenetic delayed the degeneration of articular cartilage in OA.4.3 TXC is a effective compound recipe of traditional Chinese medicine.Subject Headings:Osteoarthritis, Knee / TCD Therapy; Cartilage, Articular/ Drug effects; Controlled Clinical Trial; Rabbits @Touguxiaotong Capsule...
