Objective: To investigate the effects of Xiaoyu Jiegu powder on expression of IL-1beta,TNF-alpha and TGF-beta1in Knee Osteoarthritis(KOA) modal rabbits synovial fluid.To observe the general state of the modal rabbits, and the organization morphologicalchanges of knee joint. To provide a theoretical basis for treatment of KOA patients inclinic with Xiaoyu Jiegu powder.Methods:32Japanese white rabbits were randomly divided into4groups,8in eachgroup: the normal group, without any treatment; model group; treatment group, withXiaoyu Jiegu powder, tune with water into paste, apply evenly on the skin surface ofknee joint,30days, once a day, every eight hours; control group, with Voltaren, applyevenly on the skin surface of knee joint,30days,3times a day. Excepte normal gruop,other rabbits were fixed their left hind leg with plaster cast tube type. After6weeks,KOA model was built. After30days treatment, detect the level of IL-1beta, TNF-alphaand TGF-beta1in joint fluid of normal group and model group. Dissect the rabbit lefthind leg knee joint, to observe and capture the medial tibial plateau surface of articularcartilage. The specimens were sectioned for microscopic observation. All test data andobserve the results of single factor analysis of variance and LSD’s multiple comparisonby SPSS software,to analyze the effect of Xiaoyu Jiegu San on rabbit knee OA model.Results: Observed by gross observation and cartilage tissue sections under themicroscope, we can clearly find that, KOA model rabbits has obvious cartilagedestruction, while the pathological changes in treatment group and control group werealleviated significantly. And the effect of treatment group was better than control group.Determine the levels of IL-beta1, TNF-alpha expressed in KOA model rabbits showsthat IL-beta1, TNF-alpha was highly expressed in knee joint fluid of KOA modal rabbits,and significantly decreased after medication, the effects between treatment group and control group was not significantly different. Expression of TGF-beta1in the knee jointsshows TGF-beta1was significantly reduced in model group, and increased significantlyafter medication, and even higher than the original normal levels. The effect of twodrugs on TGF-beta1was not significantly different.Conclusion: Xiaoyu Jiegu Powder can effectively reduce the KOA cartilage damage.Through the inhibition of IL-1beta. TNF-alpha and promote the expression ofTGF-beta1in experimental rabbit knee KOA model in synovial fluid, Xiaoyu JieguPowder can play an anti-inflammatory effect on relieve and repair of cartilage damage.
