Silk Fibroin as a kind of Nature Polymer Materials has been used long before. The Waste cocoon and silk which produced in harvesting and drying silk can conserve resources, protect the environment through dealing with hot water or organic solvent and so on. Recently,the more studies with silk fibers showed them the Low immunogenicity, good biocompatibility, slow degradation rate .Research of Submandibular gland cells , Preadipocyte cells and Schwann cells which are subcutaneously injected into silks proved that cells could be attached to and extended on them and normally grown and differentiation. However, modification of silk fiber has been studied due to the poor molding as bio-materials.Nano-scale particles of silk fibroin peptide (SFP) were prepared from discarded materials of cocoon or filature by dissolving and enzymolysis. Polyvinyl Alcohol films inlaying silk fibroin peptide nano-scale particles (SFP in PVA) were prepared by blending nano-SFP and PVA in water under different blending ratios. And characterized the films and discussed the function as cell culture scaffold of bio-pharmacy technology and tissue engineering scaffold. Dissolving silk fibroin fiber in LiBr/CH3OH/H2O mixed solvent which mass ratio is 5/4/1 ,and decomposed withα-Chymotrypsin , Trypsin and Neutral, respectively. The size of SFP nano-particles prepared by neutral revealed better and appeared about 80~150nm. (SFP in PVA) Films was characterized by infrared spectroscopy (IR) measurement and shows the combination of SFP and PVA. The results of SEM showed the SFP inlayed in the PVA films with micro-segment. The surface and properties of mechanical,degradation and the biocompatibility with HepG2 and CHO cells of the (SFP in PVA) films with blending ratio of 10/90,20/80,30/70,40/60 were observed . And the results showed that SFP/PVA film with blending ratio of 30/70 has smooth surface, better mechanical properties,biodegradability and the excellent function of promoting cell growth. Therefore , it proved preliminarily the prospects for cells culture and tissue engineering scaffolds.
