Objective: Compare the different chemical composition and pharmacological action of processed Croton using traditional Chinese medicine and uyghur medicine's method, in order to discuss the processed mechanism. Methods: 1)Remove off fatty oil method and"ge li yi ke mai ti"clay method were adopted to prepare the Chinese and Uyghur's processed Croton respectively, take the content of fatty oil,moisture,ash and the rate of cribration to evaluate their qualities. 2)Use GC-MS,biurea method,ICP-AES to measure the fatty oil component,total proteins and 19 kinds of trace element respectively. 3)Discuss the drug-effect by LD50,gastrointestinal propulsive motility in mice,antiinflammatory,hemolyzation,immune function experiment. 4)According to the component result and the theroy of meridian-reaching actions to explore the change of trace element's reaching. Stepwise regression analysis was adopted to estimate the relationship between the chemical and pharmacological data. Result: 1)The processsed Croton qualities according with the《Chinese Pharmacopoeia》of 2005. 2)The fatty oil content of Croton,traditional Chinese medicine's Croton,Uyghur medicine's Croton is 58.84%,19.17%,38.80 % and GC-MS method convinced 15,17,21 kinds component ordely; the total protein concentration is 35.375 mg/g,22.395 mg/g,21.425mg/g respectively; the content of most trace element have been increased after process. 3)After Process, the toxicity,gastrointestinal propulsive motility action,antiinflammatory effect,hemolyzation action have beeen decreased sharply. Compare the two different processed Croton, the traditional Chinese medicine's Croton has higher effects on reducing the toxicity,increasing immune function,restraining hemolyzation action. 4)The unprocessed Croton mainly reaches lung and kidney, the traditional chinese medicine's Croton mainly reaches liver,spleen and heart, the uyghur medicine's Croton mainly reaches liver,spleen and kindey. The direction of trace element's reaching has been changed after process. 5) Stepwise regression analysis estimated the relationship between the chemical and the pharmacological data: Heptadecanoic acid methyl ester and LD50, 9-Octadecenoic acid-methyl ester and gastrointestinal propulsive motility action, 11-Elcosenoic acid-methyl ester and hemolyzation action, Mg and antiinflammatory effect, 9-Octadecenoic acid-methyl ester,Zn,Fe and immune function. Conclusions: 1)The processed Croton's toxicity decreased, at the same time increased the safety of clinic. 2)By process reduced the fatty oil,protein's concentration,hemolyzation action, relaxed the gastrointestinal propulsive motility action and increased the content of trace element, thereinto the traditional chinese medicine's Croton increased the immune function. 3) According to the component analysis and stepwise regression analysis we have explored the processed Croton's mechanism.
