Objective: Laparoscopic radical resection of colorectal cancer is widely practiced in the fields of general surgery. And it has been proved that laparoscopic radical resection of colorectal cancer had at least same effects of cancer treatment comparing with open surgery, with less pain, faster recovery, little interference of physiological function and lower coincidence. The success of colorectal cancer laparoscopic radical operation greatly inspires the attempt of laparoscopic radical gastrectomy. So domestic laparoscopic radical gastrectomy is practiced extensively and show promised recent effects, even most operations were done on patients with cancer in early stage. But some investigations revealed that CO2 pneumoperitoneum could boost the invasion and metastasis of cancer cells,that resulted in some worriments of application of laparoscope in cancer. It will be of great importance to investigate the relationship of carbon dioxide pneumoperitoneum and gastric cancer cellular proliferation and invasion.Methods: Different groups of cultured human gastric cancer strain SGC-7901were exposed under in vivo carbon dioxide pneumoperitoneum. model: The nude mice tumor models were built with the cancer cells by injection into abdomen and the CO2 pnemumop- eritoneum pressure was 5mmHg and 8mmHg,and the treatment time was 1h ;The Control group (the laparotomy group) underwent a midline incision,which was closed after 1h. The nude mice were be observed extensively. The expression of HPA and VEGF in tumor tissue were used to investigate the influence on proliferation and invasion ability of gastric cancer cell by RT-PCR or immunohistochemical assay.Results: The number of abdominal cavity nodules in each group was no significantly different (control group: 7.4±2.5, 5 mmHg group: 7.3±2.4, 8 mmHg group: 7.5±2.1, P>0.05). Also the total weight, the mean max diameters and distributing ranges of metastasis showed no difference in each group(P>0.05). The HPA and VEGF expression in gastric cancer tissues showed no significantly difference in each subgroup(P>0.05).Conclusion: CO2 pnemumoperitoneum did not accelerate the SGC-7901 cells proliferation ,invasion and metastasis in vivo,also it did not increase the HPA and VEGF expression。...
