ObjectivesTo study the effect of somatosatin analogue octreotide at different concentrations on proliferation in colonic cancer cell line; Through study the effect of octreotide on expression of the Wnt signal transduction pathwayβ-catenin/TCF and the downstream genes C-myc and CyclinD1 in colonic cancer cell line,to explore the effect and mechanism of action of octreotide to Wnt/β-Catenin signal pathway, to elucidate furtherly the anti-tumor mechanism of somatostatin.MethodsHuman colonic cancer cell line SW480 was studied in the experiment. MTT assay was used to analyse the effect of octreotide at different concentrations on the proliferation of SW480 cell. Western blot assay was used to evaluate the protein level ofβ-catenin,Tcf-4 and the wnt pathway downstream genes C-myc and CyclinD1 in SW480 cell treated by different concentrations of octreotide .Results1.Assays in vitro showed that octreotide inhibited proliferation of colonic cancer cell line SW480 in a dose-dependent manner in a range from 10-10 M to 10-8M and in a time-dependent manner within 48 hours. The inhibitory effect of octreotide at 10-8M was the most significant(p<0.01). Octreotide below 10-11 M had no effect on the growth of colonic cancer cell line SW480.2. The Western blot method results indicated that the protein expression ofβ-Catenin and Tcf-4 was reduced in treated SW480 cells line(OCT 10-6,10-8,10-10M).(P<0.01).3. The Western blot method results indicated that the protein expression of C-myc and CyclinD1 was reduced in treated SW480 cells line(OCT 10-6,10-8,10-10M). (P<0.01).Conclusion1. Octreotide can inhibit proliferation of colonic cancer cell line effectively in a dose-dependent and time-dependent manner in a certain degree.2.Octreotide can inhibit the amount ofβ-catenin and Tcf-4 and the wnt pathway downstream genes C-myc and CyclinD1 protein expression in SW480 cell.3.Qtreotide can suppress Wnt/β-Catenin signal transduction pathway. Octreotide suppresses Wnt/β-Catenin signal transduction pathway by its preventedβ-Catenin to enter the nucleus, inhibit the action of TCF-4 ,and thus inhibit downstream genes C-myc and CyclinD1 protein expression.
