The Research Of Intervention With Interleukin-10 On Experimental Autoimmune Thyroiditis In Rats

Objective The research of treatment with Interleukin-10 (IL-10) on experimental autoimmune thyroiditis (EAT) in Rats.To investigate the relationship between the Thyroid autoantibodies, Cytokines and the Autoimmune thyroid disease (AITD) and to investigate the treatment effects of IL-10 on the autoimmune thyroid disease.Methods 36 female SD rats were randomly divided into three groups:normal control(A), EAT control (B), Drug intervention treatment (C). n=12. Group A was injected with distilled water; Group B was injected with distilled water in Day 10 after the first immunization; Group C was injected with IL-10 in Day 10 after the first immunization. Before and after experiment:the levels of Thyroid function IL-10,Thyroid autoantibodies (TgAb and TmAb) were detected and observing pathological changes in thyroid tissue.Results 1.In group A, we see thyroid follicular mature, and no infiltration of inflammatory cells; The thyroid in group B and C could see inflammatory response and inflammatory cell infiltration; but the response in group C was weaker than group B.2.The levels of TgAb and TmAb in group B compared with Group A was statistically significant (P<0.05). The levels of TgAb and TmAb in group C was also increase, but lower than group B (P<0.05).Group B compared with Group A was statistically significant (P<0.05).The levels of IL-10 in group B was lower than in group A and group C(P<0.05). Group C compared with Group A was statistically significant (P<0.05).Conclusion 1.IL-10 can inhibit the production of inflammation, lymphocyte infiltration, autoabtibodies in thyroid.2.The levels of TgAb,TmAb changes in different stages of the AITD.They are an important symbol of the EAT immune disorders and thyroid autoimmune.3.IL-10 has positive significance in protect immune regulation of HT or EAT and has a positive treatment effect to AITD.
