Expression of IL-6,IL-8,TNF-αwas detected by reverse transcription and quantitative real time polymerase chain reaction (SYBR Greenâ… Real time -PCR).And the invasion ability of MDA-MB-231 cells was tested by Transwell chamber assay. Results:Compared with the control group,the UTI significantly inhibited the proliferation of MDA-MB-231 cells induced.The expression of IL-6,IL-8,TNF-αin MDA-MB-231 cells was down-regulated and the invasion ability of which was decreased after the treatment.Compared with the TXT group,the inhibition effect on the proliferation was weaker,but the inhibitory efficient of UTI combined with TXT was higher than that of TXT used alone.Conclusions:UTI has a synergetic effect on cell apoptosis induced by TXT in breast cancer cell line MDA-MB-231,the down-regulation IL-6,IL-8,TNF-αof expression may be one of the mechanisms. PART 2 EFFECT OF ULINASTATIN AND TAXOTERE ON INTERLEUKIN-6,INTERLEUKIN-8,TUMOR NECROSIS FACTOR-αEXPRESSION AND INHIBITION lN NUDE MOUSE WlTH MDA-MB-231 HUMAN BREAST CANCERObjective:To investigate the expression and the inhibition of Ulinastatin and Taxotere on interleukin-6,interleukin-8,tumor necrosis factor-αin nude mouse with MDA-MB-231 human breast cancer.Methods:To implantate MDA-MB-231 cell in the right armpit subcu- taneous fat pad of BALB/c-nu/nu female nude mice.There were fifty mice divided into four groups with random.There were physiological saline group,UTI group,TXT group,UTI+TXT group.They all started injecting into abdominal cavity at the three weeks. The volume of primary tumor was established by the 21th day after tumor cell inoculation. Expression of IL-6,IL-8,TNF-αwas detected by immunity class. Results:Expression of IL-6,IL-8,TNF-αprotein,that of UTI and TXT compared with control group decreased(P<0.05),UTI+TXT comp- ared with TXT,Expression of protein decreased(P<0.05),and UTI,TXT compared with control group,expression of IL-6,IL-8,TNF-αgen decreased (P<0.05),UTI+TXT compared with TXT,expression of IL-6,IL-8,TNF-αgene decreased too(P<0.05).Conclusion:Both UTI and TXT can inhibit the proliferation of breast cancer and they had synergistic effect.The mechanism may be concerned with the descending expression of IL-6,IL-8,TNF-αgene and protein by UTI.
