The ingredients profiling of seized methamphetamine samples and foreign commercial methamphetamine pseudo drug can provide very useful information to develop the Chinese own pseudo drugs in the training of narcotic dogs. The aim of this study is to develop an analytical method for detecting and identifying ingredients such as starting materials and by-products in the methamphetamine hydrochloride sample and the ingredients in the foreign commercial pseudo drug. A 50 mg methamphetamine hydrochloride sample was dissolved in phosphate buffer. The ingredients were extracted with 0.5 ml ethyl acetate, then took the organic phase 0.1μl to analyze by GC-MS equipped with DB-1 capillary column. 9 impurities were found. N-formylmethamphetamine and N-acetylmethamphetamine in the sample can prove the sample was synthesized by P-2-P through Leuckart method. N-ethylmethamphetamine exposed the P-2-P was yielded by benzaldehyde and nitroethane. From the foreign commercial pseudo drug, we found out 3 matters. They are 1,3,5-trioxane, myristic acid and 1,2-benzenedicarboxylic acid dibutyl ester. The results indicate that the present method would be effective for the methamphetamine hydrochloride and the foreign pseudo drug ingredients profiling and can provide crucial information to the study of Chinese own pseudo drugs.According to the result of former study, we established two preliminary methamphetamine pseudo drug formulas. The practical effect evaluation of the two formulas was done in this study by animal experiment. Forty Labrador Retriever dogs were equally divided into two groups, each group for one formula. The dogs accepted the standard narcotic dog training made by the Institute of Police Dog, the Ministry of Public Security with the pseudo drug, then their responses to the real methamphetamine were recorded. We found that the two groups can both 100% feel the odor of real methamphetamine after five times of odor intensification. In the formula a group, the dogs which can warn decidedly account for 90%, in the group b, the proportion is 85%. Analyzing by X~2 method, the difference is not significant. The two formulas can both meet the requirement of narcotic dog's training. According to the result of the former two studys, we established two preliminary methamphetamine pseudo drug formulas. The acute evaluation of the toxicity of each formula was done in this study by animal experiment through mouth ingestion. Eighteen dogs were equally divided into two batches, each batch for one formula. Every batch was then equally divided into three groups according to the different dosages: 10, 5, 1 ml/time. The pseudo drug was administered two times a day for 6 weeks. Blood samples were collected every week. The serum was then separated for observing 14 biochemical parameters. The result demonstrated that the two formulas are both poisonous to dogs at certain scale, but the toxicity of formula b is weaker than formula a. Therefore, in the practical training of narcotic dogs, if the effects of each formula are similar, using the formula b can reduce the potential acute poisoning.
