Objective:1.Establish insulin resistance(IR) rat models by feeding high fat diet.2. Observation and discussion the effect of exercise on Myosin Heavy Chain(MHC) in IR rat of Cardiomyocytes.3.Observation and discussion the effect of exercise on cardiac morphology,typeⅠandⅢcollagen in IR rats. To provide the sports and diet program of improvement and rehabilitation for the disease about IR caused by feeding high fat diet.Methods:To select 215 clean and healthy male Sprague-Dawley(SD)(purchase from B&K Universal Group Limited)which were 7 weeks ages and weighted from 206g to 257g,the paper divided them into 2 groups stochastically:A,Control,C,n=10, B,Model of Insulin Resistance,MIR,n=200.After buliding the model of IR rats then divided into 4 groups of MIR,were high fat diet control group(H),high fat diet exercise group(HE),ordinary diet group(O) and ordinary diet exercise group(OE). Exercise 8 weeks.After the high fat diet exercise group and ordinary diet exercise group finished its last exercises training,in the next morning the each groups rats were given a general anesthesia with chloralhydrate injection into the abdominal cavity,then were killed. Measure the blood glucose immediately and the insulin by radioimmunoassay.SDS-PAGE was adapt to measure the composition of cardiac myosin heavy chain isoform;HE staining to observe morphology changes of myocardial tissue;immunohistochemical to show expression of typeⅠandⅢCollagen in cardiac myocyte, and compute image analysis techniques for its quantitative analysis.Results:1.High fat diet can be established the model rat of insulin resistance. the insulin sensitivity index of MIR group was lower than the control group significantly. Through exercise and/or to change normal diet,insulin sensitivity index was significantly increased.2.The body weight and heart weight of high fat diet group was higher than control group significantly.After 8 weeks of aerobic exercise and (or) to change normal diet,the body weight and heart weight of 3 intervention groups, were significantly lower than high fat diet group, and close to the control group.HW/BW index of the ordinary diet group and the ordinary diet exercise group were significantly higher than the high fat diet group; HW/BW index of high fat diet group and control group no significant change.3.The composition of ventricular MHC isomers,β-MHC of high-fat diet group was significantly higher than other groups, a-MHC was significantly lower than other groups; the proportion of a-MHC in other intervention groups close to or higher than control group, no significant difference.4. The expression of typeⅠandⅢcollagen in myocardium,high fat diet group was significantly higher than the other three groups (other than ordinary diet group);high fat diet exercise group and ordinary diet exercise group values were close to control group, no significant difference.Conclusions:1.After high fat diet of 8 weeks to SD rats, can establish animal models of insulin resistance. Aerobic exercise and change to ordinary diet can improve the condition of IR significantly.2. The heart weight of IR rats increased significantly,aerobic exercise and/or resume ordinary diet can reduce heart weight adaptively;through aerobic exercise and resume ordiary diet, HW/BW index can restore normal significantly, but simple aerobic exercise no changes distinctly.3.The isoforms of MHC in Ventricular Myocardium of Insulin resistance rat, transfer to the embryo,α-MHC was increased by aerobic exercise and/or return to ordinary diet significantly, improve the heart's function.4.TypeⅠandⅢcollagen hyperplasia overly in myocardial interstitial of insulin resistance rat,a tendency to fibrosis. Aerobic exercise can reduce typeⅠandⅢcollagen in myocardial of rat,but ineffective to improve the myocardial fibrosis of simply return to ordinary diet.
