Objective: 120 one month-old healthy male SPF SD rats were selected to study through the implementation of high-fat diet in rats fed model of insulin resistance approach to building the body, and through aerobic exercise training them to observe the insulin resistance in rat myocardial tissue Expression of glucose transporter protein 4 and some related biochemical changes, designed to explore the improvement of aerobic exercise training on myocardial glucose metabolism in insulin-resistant rats the active role and importance of clinical prevention and treatment of insulin resistance broaden thinking, improving methods provide theoretical support and animal experimental evidence.Research Methods: 120 one month-old healthy male SPF SD rats were randomly divided into normal control group (NC), normal swimming exercise group (NE), IR control group (IC) and the IR swimming exercise group (IE). Respectively, the NC group and the NE group with standard feed, right IC group and the IE group with high fat diet, raising a total of 15 weeks for the construction of animal models of insulin resistance. The NE and IE groups, the second week of a period of 14 weeks of moderate-intensity aerobic endurance swimming. And the end of the cycle before the experiment testing the success of modeling, and after the execution derived using automatic biochemical analyzer were measured serum total cholesterol, triglycerides, free fatty acids and other biochemical indicators, using Western Blot Determination of myocardial cells in glucose transporter the activity of protein 4 expression.Results:①After 14 weeks'swimming training, NC group body weight was significantly higher than NE group (P <0.05), while the IC group was significantly higher than the IE group (P <0.01).②by measuring the FBG and FINS, ISI index derived rats. After the experiment, NC group was significantly smaller than the ISI index NE group (P <0.05), IC group was significantly smaller than the ISI index IE group (P <0.01).③IC serum TC was significantly higher than the NC group (P <0.01), IE serum TC was significantly higher than the NE group (P <0.05); IC serum TG was significantly higher than the NC group (P <0.01), IE group TG was significantly higher than NE group (P <0.01); IC serum FFA were significantly higher than NC group were significantly higher than the IE group (P <0.01).④NE serum SOD activity was significantly higher than the NC group (P <0.01), also significantly higher than the IE group (P <0.05), IE serum SOD activity was significantly higher than the IC group (P <0.01); IC group Serum MDA content was significantly higher than the NC group (P <0.01), NC was significantly higher than the NE group (P <0.05), NE was significantly higher than the IE group (P <0.01).⑤NC GLUT4 expression in myocardial tissue, significantly higher than the IC group, but also significantly higher than the NE group (P <0.01). IE group GLUT4 expression in myocardial tissue, significantly higher than the IC group (P <0.01). NE group GLUT4 expression in myocardial tissue, significantly higher than the IE group (P <0.01).Conclusion: In rat fat diet can induce insulin resistance, model construction, low cost, into the mold after the stabilization of the security. The role of oxidative stress and insulin resistance are closely related, will increase the body's oxidation is excessive lipid peroxide generation, produce damage to the body tissue. Aerobic exercise training can to some extent mitigate the occurrence and development of this trend, maintaining the body's internal balance. In addition, the high-fat diet induced insulin resistance, inhibit the activity of glucose transporter 4 expression, but also to increased serum free fatty acid levels, leading to increased fat on myocardial function and myocardial cells reduced capacity to use glucose and produce toxic substances affect the lipid Myocardial overall function. Aerobic exercise training can reduce levels of serum free fatty acids to alleviate the situation of insulin resistance, stimulate cardiac cells the activity of glucose transporter 4 expression and promote a state of insulin resistance in glucose metabolism in the body function in normal operation to improve the body's insulin sensitivity.
