The amino acid neurotransmitters are classified into EAA and IAA. EAA includes Glu and Asp, and IAA includes GABA and Gly. Now Pro is also considered as inhibitory amino acid neurotransmitter. A large of studies show that in normal state the contents of EAA and IAA are relatively balanced, and the metabolism of Glu and GABA is also relatively balanced. Both Glu and GABA are normal metabolic offspring for cerebrum. But when the body is fatigued with exercise, the amplitude of IAA is higher than that of EAA. This situation leads to the cerebrum inhibition prioring to its excitability. The other studies show the increase of GABA, as one of the key factor induced central fatigue, is caused by the decrease of oxidation. That is, the activity of GABA-T SSADH is decreased and the activity of GAD is increased. GABA s lysosomal is handicapped which leads to GABA s accumulation in cerebrum. The Chinese medicine semen astragali complanati contains many effective components, such as amino acid, peptide, proteins, falconoid, trepans and organic acids, which can increase the flow of the blood in brain, decrease lipids and protect liver, etc. Recently semen astragali complanati is processed into many healthy foods. The objective of this experiment is to study the gene expression of GAD67, GABA-T, SSADH and the effects of semen astragali complanati on them and the contents of the amino acid neurotransmitters in different brain region of exercise-induced central fatigue rats. These results can provide experimental basis for discussing the mechanisms of exercise-induced central fatigue and exploring semen astragali complanati as sports healthy food.In this experiment, 48 male SD rats were randomly divided into four groups, and seven weeks made the sport fatigue model. All rats were killed immediately after exercise at the start of eight week, and the rest control group was killed in quiet state at the same time. Then the contents of amino acid neurotransmitters were tested by the amino acid automatic analytical instrument in hippocampus and diencephalons of all rats; and the gene expression of GAD67 GABA-T and SSADH were determine by RT-PCR technique in the same region.The main results were as follows:l)Semen astragali complanati could efficiently inhibited the decrease of rat's weight caused by the heavy exercise. It suggested that semen astragali complanati could enhance substance metabolism that was good for promoting the utilization and storage of energy during exercise and body's recovery after exercise.2)The gene expression of GAD67 mRNA in cerebrum of exercise-induced fatigue rat was increased significantly(P<0.05). It indicated the reason of the change of GAD's activity in cerebrum of exercise-induced fatigue rat; Semen astragali complanati could decrease the gene expression of GAD67 mRNA by bettering the state for lack of oxygen in cerebrum.3)The gene expression of GABA-T SSADH were declined significantly(P<0.05) in different brain region of exercise-induced fatigue rat. It indicated the reason of the change of GABA-T SSADH activity in cerebrum of exercise-induced fatigue rat. Semen astragali complanati could strengthen the gene expression of GABA-T SSADH by modifying the body's equilibrium state.4) Semen astragali complanati could regulate the level of Gku GAB A in different brain region of exercise-induced fatigue rat. At the same time the level of Asp Gly Pro were regulated. It showed semen astragali complanati could regulate the stability of body's internal environment and promote the balance of excitant state and inhibitory state in cerebrum.5) Semen astragali complanati increased the ratio of Glu/GABA in cerebrum of exercise-induced fatigue rat and maintained the ratio to a normal level, which showed semen astragali complanati could modify the metabolism equilibrium of Gku GAB A, so that enhanced the excitability of nervous system.6) Semen astragali complanati could reduce the level of L-Arg in cerebrum of exercise-induced fatigue rat, which suggested it could prevent the excess synthesis of NO.The above results...
