Objects:This research aimed at discussing the relationship between hepatocyte' s apoptosis and middle intensity exercise of different time through comparing apoptosis, liver organization glycogenic content, NO content, expresssion of Bcl-2 and Bax of rats.Methods:30 10-week age healthy grown-up male SD rats were divided into 5 groups at random: Comparison Group (Control, C. No exercise);Middle Intensity Exercise Group:Twenty-Minute -Exercise Group(TME);One-Hour-Exercise Group (OHE);Two-Hour- Exercise Group(THE);Exhausting Exercise Group ( EE ) .The middle intensity exercise model of rats according as Bedford exercises at the speed of 19. 3 m ? min-1. All rats were broken the neck after the anaesthesia on the berry to fetch the liver organization, a part was used to test the organization glycogenic content and NO content, another part to prepar paraffin wax section and to measure hepatocyte apoptotic and the expression of Bcl2 and Bax protein.Results: (1) The slice was showed in HE dyeing:In the C Group and the TME Group, the structures of rats' liver cell were completed, core big and circular;In the groups of OHE and THE, liver cell structure of rats changed gradually, and the liver cell of the EE group began to be obviously swelling, the liver cell gap was obviously enlarged, the core became small and irregular and hepatic lobe outline changed. (2) The TUNEL dyeing slice showed that male-dyeing grains of hepatocyte' s apoptosis increased gradually in rats with extension of time. Statistic results showed: hepatocyte's AI had no statistic meaning among the C, TME and OHE group (p > 0. 05) . Conversely, among the Exercise Groups, there was a significant difference (p< 0. 01). When the OHE, THE and EE group were each compared, the apototic of the EE group was the highest.(3) With extension of time in every group, glycogen content dropped accordingly. When it exercised to exhaust, glycogen content closed to wear out. There was no statistic meaning between the C and TME group (p> 0. 05);When compared the rest exercise groups with the C group, we could found there was a significant difference (jcKO. 01 );Among exercise groups, there was a highly significant difference (p< 0.01) between the rest exercise groups in the aspect of glycogen content except that there was no statistic meaning between the THE and EE group (p>0. 05) .(4) When the rats of the TME, OHE group were compared with those of the C group, NO content dropped and the difference had no statistic meaning (p>0. 05) .When the rats of the OHE, THE and EE groups were compared with each other, the difference of NO content' s dropping was remarkable, the difference of NO content between the OHE and EE Group was highly remarkable(p< 0. 01).(5) The dyeing slice of Bcl-2 and Bax showed:With the extension of time, male-dyeing grains increased gradually and the grey degree was strengthened. The statistic results showed that there was no statistic meaning between the C group and the Exercise group (the TME and OHE groups) in Bcl-2 and Bax's expressions (p>0. 05) .There was also no highly significant meaning among the Exercise groups(the OHE, THE and EE Group) (p< 0. 01). The Bax/Bcl-2 value showed:The value became larger in the C group , TME , OHE and THE group and the difference had no statistic meaning between them (p > 0.05) .There was a significant difference when the EE group was compared with the other groups in Bax/Bcl-2 value.(6) The relations between hepatocyte apoptosis and these items showed that there was a highly significant negative co-rrelation between AI and glycogen , NO content and a highly significant positive co-rrelation between AI and Bcl-2, Bax (p< 0. 01).Conclusion:(1)It had no significant effect on hepatocyteapoptosis of rats after 20min or lh middle intensity exercise.(2)2h middle intensity exercise may cause hepatocyte apoptosis of rats, and the apoptotic was more obvious when they exercised to exhaust.(3)The glycogen and No content had negative relativity with hepatocyte apoptosis of rats.(4) Bcl-2 and Bax participated in regulating the hypatocyte' s apoptosis, and the balance relationship between the two were important factors if they would maintain or induce apoptosis.
