[ Objective ]1, Establish a sport rat model of jump , provide a valid reference for studying the influence of the sport model of jump on BMD and bone metabolism and the internality mechanism;2, Study the effects of the jump model on BMD in the rat;3, Study the effects of the jump exercise and the swimming exercise on the biochemical indexes of bone metabolism;4, Study the effects of the jump on the expression of BMP-2 mRNA in the rat;The final purpose of this paper is to explore the effects and mechanisms of jump on bone metabolism in the growing rat .Compare with the swimming exercise and analyze the difference of the bone metabolism caused by two kinds of sport modes. Provide the reference for the teenager to choose the reasonable sport mode to improve their health. Provide the reasonable theory for the research preventing the osteoporosis in the old people. [ Method ]1, Establish an animal jump sport model: Make the box of jumping for rat. Put the rat into the box of jumping, and switch electricity on, the rat will jump on the safe terrace under the electric stimulation, then take a 20-second rest. Doing it repeatedly until complete the training.2, Observation on the rat model: Observe the activities, diet, body weight, color and luster of rat's hair during experiment. After the last exercise, take the blood of rat tail, and measure the BLA by the automatic lactic acid analyzer;3 , Measure biochemical indexes of bone metabolism: Serum Ca, P, ALP and TRACP were measured by 721 Spectrometrycents.4, Basic physical items of tibiae and femur of left: Measure tibiae and femur as following index: thickness and width of outlying, the thickness and width of other end and length;5, Measurement BMD of the rat's femur of left: Measure bone mineral content and BMD by dual energy X-ray absorptiometry (DEXA).6, Measure the level of BMP-2 mRNA in femur cartilage tissue : Measure the level of BMP-2 mRNA in femur cartilage tissue by the technique of RT- PCR. And compute the optical-density indexes of BMP-2 mRNA in each sample, account the ratio with the optical-density indexes of GAPDH by analytical system. [ Result ]1, The observation of the rat's basic circumstance: During experiment rat moved and ate freely. The jump group has no difference on the weight compared with the control. The swimming group is lighter than the former on the weight, but the difference is not obvious ( P>0.05).2, Compare the modality indexes of femur and tibiae in the rat: Although the jump group and the swimming group show the increased trend on the modality indexes compared with the control, the difference is not obvious (P>0.05).3, Compare the BMD of femur in the rat: The jump group has the obvious difference compared with the control (P<0.05); although the swimming group shows the increased trend on BMD compared with the control, the difference is not obvious (P>0.05).4, Compare the BMD of vertebra in the Tat: The jump group has the obvious difference compared with the control (P<0.05); the swimming group has the remarkable difference compared with the control (P<0.01).5, Compare the level of BLA in the rat: The jump group has the obvious difference on the level of BLA compared with the control (P<0.05); the swimming group has the remarkable difference on the level of BLA compared with the control(P<0.05).6, Compare the level of Serum Ca and P in the rat: The jump group and the swimming group have no obvious difference on the level of Serum Ca and P compared with the control( P>0.05).7, Compare the level of ALP in the rat: The jump group has remarkable difference on the level of ALP compared with the control (P<0.01); the swimming group has the obvious difference on the level of ALP compared with the control (P<0.05).8, Compare the level of TRACP in the rat: The jump group and the swimming group have the obvious difference on the level of TRACP compared with the control (P<0.05).9, Compare the level of BMP-2 mRNA in femur cartilage tissue: The jump group has remarkable increase on the level of BMP-2 mRNA compared with the control (P<0.01); the swimming group has obvious increase on the level of BMP-2 mRNA compared with the control (P<0.05). [ Conclusion ]1, Establish animal jump sport model in this experiment. Successfully simulate the main physiology stimulation brought by the jump on bone metabolism of the rat, and provide the preferable animal model for the further research on the inference of the jump sport on the body.2, After 8-week exercise, The biochemical indexes of bone metabolism show that jump can promote bone growing effectively compared with swimming.3, The sport can effectively promote the growth of bone. The BMD of femur and vertebra increased obviously. The influence of jump is more obvious than swimming's on BMD of femur.
