Through studying the effects of a bout of aerobic exercise onblood sugar and p38 in skeletal muscle of KK-AyⅡdiabetes mice,the thesis will discuss possible mechanism of glucose-loweringthrough exercise from p38MAPK pathways and then provides thescientific basis and the reference for cure diabetes by exercise.Six-month-old male KK-Ay & C57BL/6J mouse each 12, were dividedinto 4 groups randomly according to the blood sugar: diabetesexercise group (KE group) and diabetes control group (KC group);normal exercise group (CE group) and normal control group (CC group).Exercise project: 0°grade; speed in 12m·min-1; 30min. Blood andthe quadriceps femoris muscle were obtained from the mice immediatelyafter the experiment, before that anaesthesia was executed. Determination the blood sugar levels with GOF-PAP. Detection p38 proteinexpression and phosphorylation by western blot. Determination bloodserum free fatty acids in improvement color method. Test seruminsulin and TNF-αthrough radioimmunoassay.Results: Body weight of KK-Ay mouse was higher than theC57BL/6J's. Blood sugar of KK-Ay mouse was higher than theC57BL/6J's. Blood sugar was reduced in KE group and CE group aftera bout of acute exercise. Blood serum insulin levels in KC group is 59-fold of CC group. It was 55%lower in CE group than in CC group.It was 43%lower in KE group than in KC group, p38 phosphorylationof KC group was increased than CE group's in skeletal muscle. Itwas increased in CE group than in CC group. And increased in KE groupthan in KC group also. The difference above has statisticssignificance. Serum TNF—αof KC group was higher than the CC groupobviously. It was lower in KE than in KC group. Blood serum FFA washigher in KC than in CC group obviously. It was lower in KE than inKC group. Although it was lower in CE than in CC group, the differencehas no statistics significance.Conclusion: p38MAPK phosphorylation was increased in skeletalmuscle of KK-Ay mice. Levels of serum TNF-αand FFA was increasedat the same time. Aberrant p38 MAPK signalling might contribute tothe pathogenesis of insulin resistance ofⅡdiabetes, p38 and TNF-α&FFA effect on each other. All of these play the extremely vitalrole in insulin resistance andⅡdiabetes developing processtogether. Firstly, a bout of aerobic exercise effect on p38 inskeletal muscle and serum TNF-α&FFA of KK-Ay mouse directly: theactivation of p38 phosphorylation in skeletal muscle; reduction oflevels of serum TNF-αand FFA. Then indirect effect on TNF-αand FFAthrough the p38. Finally reduces the blood sugar levels. This ispartial mechanisms of glucose-lowering through exercise.
